microbacterium ginsengiterrae sp. nov., a beta-glucosidase-producing bacterium isolated from soil of a ginseng field.strain dcy37(t) was isolated from a soil sample of a ginseng field in the republic of korea and characterized in order to determine its taxonomic position. cells were gram-staining-positive, heterotrophic, strictly aerobic, non-motile short rods. 16s rrna gene sequence analysis revealed that strain dcy37(t) belongs to the genus microbacterium. according to 16s rrna gene sequence analysis, it is closely related to microbacterium aerolatum dsm 14217(t) (98.8 %), microbacterium hydrocarbonoxydans d ...201020081025
characterization of a recombinant (-)γ-lactamase from microbacterium hydrocarbonoxydans.a (-)γ-lactamase, mhg, from microbacterium hydrocarbonoxydans was over-expressed in e. coli and was characterized after purification. the maximum activity was at ph 8.0 and 60°c and the half life of mhg was ~30 min at 75°c. the enzyme was activated by dtt. the catalytic triad of the (-)γ-lactamase is comprised of residues ser98, asp230, and his259 and an oxyanion hole was formed by tyr32 and met99 according to the alignment results. under native conditions, the (-)γ-lactamase consists of two 31  ...201121968479
identification and characterization of a novel (+)-γ-lactamase from microbacterium hydrocarbonoxydans.2-azabicyclo[2.2.1]hept-5-en-3-one (γ-lactam) is an important precursor of many carbocyclic nucleoside analogs and pharmaceuticals. (-)-γ-lactam has attracted much attention because of its role as an intermediate of antiviral drugs such as abacavir and carbovir. (+)-γ-lactamase can be used for the kinetic resolution of γ-lactam to obtain (-)-γ-lactam. in this study, a novel (+)-γ-lactamase (mh33h4-5540) was discovered from the gene library of microbacterium hydrocarbonoxydans based on a colorime ...201627255489
Microbacterium murale sp. nov., isolated from an indoor wall.A Gram-positive rod shaped bacterium (01-GI-001T) isolated from a wall colonized with moulds was studied for its taxonomic position. The 16S rRNA gene sequence analysis showed, that the isolate clearly grouped into the Microbacterium cluster. On the basis of pairwise comparisons of the 16S rRNA gene sequences, isolate 01-Gi-001T was most closely related to the type strains of Microbacterium hydrocarbonoxydans (98.8%), Microbacterium profundi (98.7%), Microbacterium phyllosphaerae (98.3%) and Mic ...201122199224
enantioselective resolution of γ-lactam by a whole cell of microbacterium hydrocarbonoxydans (l29-9) immobilized in polymer of pva-alginate-boric acid.using immobilized cells of a novel strain of microbacterium hydrocarbonoxydans l29-9 in polymers of polyvinyl alcohol (pva)-alginate-boric acid, enantioselective resolution of racemic γ-lactam to produce (-)γ-lactam was successfully carried out. a 6:1 ratio of pva:sodium alginate not only prevented agglomeration of the matrix but also produced beads with high gel strength. the optimum biotransformation conditions were 1 g/l substrate, ph 7.0, reaction temperature of 30 °c, and reaction time of 3 ...201020563863
microbacterium oleivorans sp. nov. and microbacterium hydrocarbonoxydans sp. nov., novel crude-oil-degrading gram-positive bacteria.a taxonomic study of two crude-oil-degrading, gram-positive bacterial strains, designated bas69(t) and bnp48(t), revealed that they represent two novel microbacterium species. 16s rrna gene sequence similarity to their closest phylogenetic neighbours was 98.5 % for bas69(t) (microbacterium paraoxydans dsm 15019(t) and microbacterium saperdae dsm 20169(t)) and 99 % for bnp48(t) (microbacterium luteolum dsm 20143(t)). levels of dna-dna relatedness to the closest phylogenetic neighbours of both str ...200515774639
characterization of a recombinant (+)-γ-lactamase from microbacterium hydrocarbonoxydans which provides evidence that two enantiocomplementary γ-lactamases are in the strain.a two-step method, i.e., the transfer acyl analysis and then the chiral hplc analysis, was employed in the screening of the cosmid library of microbacterium hydrocarbonoxydans genome. two enantiocomplementary γ-lactamase clones were found. a 40-kb cosmid showed (-)-γ-lactamase activity, and the activity was from mhg which was reported previously according to the results of pcr identifying experiment. the 37-kb (+)-γ-lactamase cosmid was further constructed into a puc18 plasmid library and screen ...201525341397
identification and regulation of the catalytic promiscuity of (-)-γ-lactamase from microbacterium hydrocarbonoxydans.mhg, a previously reported (-)-γ-lactamase from microbacterium hydrocarbonoxydans, was identified to have perhydrolase activity by combining structure similarity search with activity assays. kinetic studies illustrated that perhydrolysis was the native activity owing to lower k m and higher k cat/k m values. experimental evidence showed that both hydrolysis and perhydrolysis reactions took place at the same active center. engineering of the putative substrate-binding pocket revealed that leu233 ...201525773976
structural insights into the γ-lactamase activity and substrate enantioselectivity of an isochorismatase-like hydrolase from microbacterium hydrocarbonoxydans.(+)-γ-lactamase catalyzes the specific hydrolysis of (+)-γ-lactam out of the racemic γ-lactam (2-azabicyclo[2.2.1]hept-5-en-3-one) to leave optically pure (-)-γ-lactam, which is the key building block of antiviral drugs such as carbovir and abacavir. however, no structural data has been reported on how the enzymes bind the γ-lactams and achieve their enantioselectivities. we previously identified an isochorismatase-like hydrolase (ihl, mh33h4-5540) with (+)-γ-lactamase activity, which constitute ...201728295028
microbacterium arthrosphaerae sp. nov., isolated from the faeces of the pill millipede arthrosphaera magna attems.a gram-reaction-positive, rod-shaped bacterium, designated strain cc-vm-y(t), was isolated from the faeces of the pill millipede arthrosphaera magna attems from india and was subsequently studied to determine its taxonomic position. based on16s rrna gene sequence similarities, the isolate clearly grouped with members of the genus microbacterium. on the basis of pairwise comparisons of the 16s rrna gene sequences, strain cc-vm-y(t) was most closely related to microbacterium insulae ds-66(t) (98 % ...201020656817
