isolation and characterization of adeno-associated viruses from bovine adenovirus types 1 and 2.hemagglutinating dna viruses of 20 nm diameter were isolated from bovine adenovirus types 1 and 2. the isolates were heat stable, chloroform resistant, and defective. their densities were 1.38 to 1.39 g/cm3, and they were found to be serologically identical to the bovine adeno-associated virus strain x7. a partial antigenic relationship was found between these and the canine adeno-associated virus.1976986122
cloning and characterization of a bovine adeno-associated virus.to better understand the relationship between primate adeno-associated viruses (aavs) and those of other mammals, we have cloned and sequenced the genome of an aav found as a contaminant in two isolates of bovine adenovirus that was reported to be serologically distinct from primate aavs. the bovine aav (baav) genome has 4,693 bp, and its organization is similar to that of other aav isolates. the left-hand open reading frame (orf) and both inverted terminal repeats (itrs) have the highest homolo ...200415163744
expression profiles of bovine adeno-associated virus and avian adeno-associated virus display significant similarity to that of adeno-associated virus type 5.we present the first detailed expression profiles of nonprimate-derived adeno-associated viruses, namely, bovine adeno-associated virus (b-aav) and avian adeno-associated virus (a-aav), which were obtained after the infection of cell lines derived from their natural hosts. in general, the profiles of b-aav and a-aav were quite similar to that of aav5; however, both exhibited features found for aav2 as well. like adeno-associated virus type 5 (aav5), b-aav and a-aav utilized an internal polyadeny ...200616699028
gangliosides are essential for bovine adeno-associated virus entry.recombinant adeno-associated viruses (aav) are promising gene therapy vectors. we have recently identified a bovine adeno-associated virus (baav) that demonstrates unique tropism and transduction activity compared to primate aavs. to better understand the entry pathway and cell tropism of baav, we have characterized the initial cell surface interactions required for transduction with baav vectors. like a number of aavs, baav requires cell surface sialic acid groups for transduction and virus att ...200616699032
the transcription strategy of bovine adeno-associated virus (b-aav) combines features of both adeno-associated virus type 2 (aav2) and type 5 (aav5).the parvoviruses bovine adeno-associated virus (b-aav) and adeno-associated virus type 5 (aav5) have similar transcription maps. however, while the aav5 capsid gene promoter p41 possesses a high basal level in 293 cells, and is further activated only poorly by rep during adenovirus type 5 (ad5) infection, the b-aav p41 promoter has a low basal activity within repcap constructs in these cells and can be strongly activated by its rep protein in the presence of ad5 when a rep-binding element (rbe) ...200817939931
coordinated control of connexin 26 and connexin 30 at the regulatory and functional level in the inner ear.connexin 26 (cx26) and connexin 30 (cx30) are encoded by two genes (gjb2 and gjb6, respectively) that are found within 50 kb in the same complex deafness locus, dfnb1. immunocytochemistry and quantitative pcr analysis of cx30 ko mouse cultures revealed that cx26 is downregulated at the protein level and at the mrna level in nonsensory cells located between outer hair cells and the stria vascularis. to explore connexin coregulation, we manipulated gene expression using the bovine adeno-associated ...200819047647
transduction of rat pancreatic islets with pseudotyped adeno-associated virus vectors.pancreatic islet transplantation is a promising treatment for type i diabetes mellitus, but current immunosuppressive strategies do not consistently provide long-term survival of transplanted islets. we are therefore investigating the use of adeno-associated viruses (aavs) as gene therapy vectors to transduce rat islets with immunosuppressive genes prior to transplantation into diabetic mice.200919450275
gene transfer using bovine adeno-associated virus in the guinea pig cochlea.gene transfer into the cells of the cochlea is useful for both research and therapy. bovine adeno-associated virus (baav) is a new viral vector with potential for long-term gene expression with little or no side effects. in this study, we assessed transgene expression using baav with beta-actin-gfp as a reporter gene, in the cochleae of normal and deafened guinea pigs. we used two different routes to inoculate the cochlea: scala media (sm) or scala tympani (st). auditory brainstem response asses ...200919458651
baav transcytosis requires an interaction with beta-1-4 linked- glucosamine and gp96.cell surface carbohydrates play an important role in virus entry and intracellular trafficking. bovine adeno-associated virus (baav) uses plasma membrane gangliosides for transduction and infection. in addition, independent of the infectious pathway, baav also has the ability to pass through barrier epithelia and endothelia using a transcytosis pathway dependent upon the presence of cell surface carbohydrates. thus, in order to better define the carbohydrate interactions that are necessary for b ...201020231878
viral vector tropism for supporting cells in the developing murine cochlea.gene-based therapeutics are being developed as novel treatments for genetic hearing loss. one roadblock to effective gene therapy is the identification of vectors which will safely deliver therapeutics to targeted cells. the cellular heterogeneity that exists within the cochlea makes viral tropism a vital consideration for effective inner ear gene therapy. there are compelling reasons to identify a viral vector with tropism for organ of corti supporting cells. supporting cells are the primary ex ...201121530627
BAAV mediated GJB2 gene transfer restores gap junction coupling in cochlear organotypic cultures from deaf Cx26Sox10Cre mice. The deafness locus DFNB1 contains GJB2, the gene encoding connexin26 and GJB6, encoding connexin30, which appear to be coordinately regulated in the inner ear. In this work, we investigated the expression and function of connexin26 and connexin30 from postnatal day 5 to adult age in double transgenic Cx26(Sox10Cre) mice, which we obtained by crossing connexin26 floxed mice with a deleter Sox10-Cre line. Cx26(Sox10Cre) mice presented with complete connexin26 ablation in the epithelial gap junctio ...201121876744
Bovine AAV transcytosis inhibition by tannic acid results in functional expression of CFTR in vitro and altered biodistribution in vivo.Bovine adeno-associated virus (BAAV) can enter a cell either through a transcytosis or transduction pathway. We previously demonstrated that particles entering via the transcytosis pathway can be redirected to transduce the cell by blocking particle exocytosis with tannic acid (TA). To investigate whether this approach is useful in lung gene therapy applications, we tested the effect of TA on BAAV transduction in cystic fibrosis airway epithelia in vitro, and in mouse lung in vivo. Our findings ...201122011646
a metagenomics and case-control study to identify viruses associated with bovine respiratory disease.bovine respiratory disease (brd) is a common health problem for both dairy and beef cattle, resulting in significant economic loses. in order to identify viruses associated with brd, we used a metagenomics approach to enrich and sequence viral nucleic acids in the nasal swabs of 50 young dairy cattle with symptoms of brd. following deep sequencing, de novo assembly, and translated protein sequence similarity searches, numerous known and previously uncharacterized viruses were identified. bovine ...201525740998
