aquaculture of three phyla of marine invertebrates to yield bioactive metabolites: process developments and economics.large-scale, renewable supplies of chemical constituents derived from marine invertebrates have limited development of potential new natural product drugs. this paper describes the development of two in-sea aquaculture systems designed and engineered for production of large quantities of biomass for two species of marine invertebrates desired for their natural product chemical constituents. the two invertebrates and their products were: (1) the cosmopolitan, arborescent bryozoan bugula neritina ...200312919831
antifouling substances against larvae of the barnacle balanus amphitrite from the marine sponge, acanthella cavernosa.a total of 118 japanese marine invertebrates were extracted with methanol, and their chloroform-soluble materials tested for antifouling activity as well as toxicity against barnacle (balanus amphitrite) larvae. among 86 species that showed more than 80% larval settlement inhibition, 13 species, including 8 sponges were weakly toxic to the barnacle larvae. the sponge acanthella cavernosa, which showed most promising activity, was extracted with ethanol and fractionated by solvent partitioning, s ...200314618720
phospholipase a2 in porifera.phospholipase a2 (pla2) catalytic activity was measured in aqueous extracts of 83 freeze-dried specimens representing 55 marine sponge species collected from the east coast of australia including the great barrier reef. high levels (>500 u/l) of pla2 activity (defined as the amount of activity that releases 1 micromol of fatty acid per min) were found in four out of 55 species (7%), moderate activities (100-499 u/l) in 6/55 (11%), low activities (1-99 u/l) in 11/55 (20%) and no pla2 activity in ...200415050528
antifouling properties of 10beta-formamidokalihinol-a and kalihinol a isolated from the marine sponge acanthella cavernosa.many soft-bodied sessile marine invertebrates such as sponges and soft corals defend themselves against fouling directly through the production of antifouling compounds, or indirectly through regulating the epibiotic microbes that affect larval settlement. in this study, 10beta-formamidokalihinol-a and kalihinol a were isolated and purified from the marine sponge acanthella cavernosa (dendy). the results indicated that both compounds inhibited the growth of bacteria isolated from the natural env ...200616551558
isocyano compounds as non-toxic antifoulants.the marine sponge acanthella cavernosa and nudibranchs of the family phyllidiidae contain isocyanoterpenoids and their related compounds that show potent antifouling activity against cypris larvae of the barnacle balanus amphitrite, while their toxicity to cyprids is weak. to develop non-toxic antifoulants based on isocyanoterpenoids, especially 3-isocyanotheonellin, a total of 36 isocyano compounds have been synthesized. they were evaluated by both antifouling activity and toxicity toward b. am ...200616805439
a comparison of sesquiterpene scaffolds across different populations of the tropical marine sponge acanthella cavernosa.specimens of the indo-pacific sponge acanthella cavernosa dendy collected from locations along the eastern coastline of australia have been shown to contain a range of sesquiterpenes including isothiocyanate 1, isocyanide 10, and the isocyanates 15 and 22. these metabolite studies have provided a basis for chemical comparisons between sponge populations from different geographic locations and between individual specimens collected from a single location.200717953446
new a-nor steroids and their antifouling activity from the chinese marine sponge acanthella cavernosa.a chemical examination of the chinese sponge acanthella cavernosa resulted in the isolation of three new a-ring contracted steroids, the ethyl esters of 2beta-hydroxy-4,7-diketo-a-norcholest-5-en-2-oic acid (1), 24s-ethyl-2beta-hydroxy-4,7-diketo-a-norcholest-5-en-2-oic acid (2), and 2beta-hydroxy-4,7-diketo-24r-methyl-a-norcholest-5,22(e)-dien-2-oic acid (3), along with four other known steroids (4-7). the structures were determined on the basis of extensive spectroscopic analysis. compounds 1- ...200818812182
confirmation of the configuration of 10-isothiocyanato-4-cadinene diastereomers through synthesis.the marine sponge metabolite 10-isothiocyanato-4-cadinene (1) was first isolated by garson et al. from acanthella cavernosa in 2000. the same structure 1 was later reported by wright et al. from the nudibranch phyllidiella pustulosa and its sponge diet, but with different nmr data. the syntheses of both enantiomers of 1 were accomplished through the isothiocyanation of 10-isocyano-4-cadinene (2) previously synthesized by our group. the correct spectroscopic data and specific rotation value of th ...201223163354
tricyclic guanidine alkaloids from the marine sponge acanthella cavernosa that stabilize the tumor suppressor pdcd4.a cell-based high-throughput screen that assessed the cellular stability of a tumor suppressor protein pdcd4 (programmed cell death 4) was used to identify a new guanidine-containing marine alkaloid mirabilin k (3), as well as the known compounds mirabilin g (1) and netamine m (2). the structures of these tricyclic guanidine alkaloids were established from extensive spectroscopic analyses. compounds 1 and 2 inhibited cellular degradation of pdcd4 with ec50 values of 1.8 μg/ml and 2.8 μg/ml, resp ...201425196934
in four shallow and mesophotic tropical reef sponges from guam the microbial community largely depends on host identity.sponges (phylum porifera) are important members of almost all aquatic ecosystems, and are renowned for hosting often dense and diverse microbial communities. while the specificity of the sponge microbiota seems to be closely related to host phylogeny, the environmental factors that could shape differences within local sponge-specific communities remain less understood. on tropical coral reefs, sponge habitats can span from shallow areas to deeper, mesophotic sites. these habitats differ in terms ...201627114882
formamido-diterpenes from the south china sea sponge acanthella cavernosa.seven new formamido-diterpenes, cavernenes a-d (1-4), kalihinenes e and f (5-6), and kalihipyran c (7), together with five known compounds (8-12), were isolated from the south china sea sponge acanthella cavernosa. structures were established using ir, hresims, 1d and 2d nmr, and single x-ray diffraction techniques. the isolated compounds were assessed for their cytotoxicity against a small panel of human cancer cell lines (hct-116, a549, hela, qgy-7701, and mda-mb-231) with ic(50) values in the ...201222851918
new diterpene isonitriles from the sponge phakellia pulcherrima.seven new diterpene isonitriles and isothiocyanates were isolated from the sponge phakellia pulcherrima along with eight known ones. six of the new compounds, 9-14, and the eight known ones, 1-8, belong to the kalihinol family of diterpenes. structures were determined from spectroscopic data. this is the first report of diterpenes from sponges of this genus. all of the other kalihinol diterpenoids have been isolated from the sponge acanthella cavernosa collected in diverse locations.19989868156
six new diterpene isonitriles from the sponge acanthella cavernosa.six new diterpene isonitriles, 10-epi-isokalihinol f [1], 10-epi-isokalihinol h [2], 1-epi-kalihinene [4], 15-isothiocyanato-1-epi-kalihinene [5], 1,10-diepi-kalihinene [6], and kalihipyran [7] were isolated, together with the known compound isokalihinol b [3], from acanthella cavernosa collected in the seychelles. the structures of the new diterpenes were elucidated by interpretation of spectral data. the ring system of kalihipyran [7] differs from that of the kalihinols and kalihinenes.19948021650
