the role of mite allergen in chronic urticaria.fifty-two patients with chronic urticaria were tested intradermally with mite allergen (dermatophagoides farinae). none had any past history of any other atopic diseases. thirty of the 52 patients gave positive skin tests to mite allergen. a good correlation between skin sensitivity to mite allergen and in vitro mite antigen-induced histamine release from leucocytes was observed. these facts suggest that a mite may be an important role in chronic urticaria.197992903
clinical correlations between long-term (ige) and short-term (igg s-ts) anaphylactic antibodies in atopic and 'non-atopic' subjects with respiratory allergic disease.the sera of atopic and non-atopic persons with allergic pulmonary disorders were examined for long-term sensitizing, ige and short-term sensitizing heat-stable (s-ts) antibodies which were present separately or together in the sera of some patients sensitive to antigens such as budgerigar serum proteins and aspergillus funigatus. in fourteen atopic patients with extrinsic asthma, six had both types of antibody to common allergens, and of nine non-atopic patients with crytogenic (intrinsic) asthm ...1979117951
characterization of the allergenic components of the house dust mite, dermatophagoides farinae.characterization and purification of the allergenic components of the house dust mite, dermatophagoides farinae, were investigated. it was found that the mite antigen was heat-stable, pronase-sensitive, and periodate-resistant, which suggested that the antigenic determinants of the mite antigen resided mainly in the protein component. purification of the mite antigen was tried using sephadex g-150, deae cellulose, and isoelectrofocusing columns. the most potent allergenic fraction was around 20, ...1977201571
specific ige estimations by rast in japanese asthmatics compared with skin, passive transfer and bronchial provocation tests.198 asthmatics and twenty healthy persons were studied by rast and in vivo tests with four common inhalant allergens. (a) higher rast classes were elicited with mite (dermatophagoides farinae) extract and lower classes with other allergens. the agreement between positive rast and skin or p-k tests was highest with the mite extract. rast sensitivity was dependent on the kinds of allergens and was most sensitive to mite extract. positive rast was most closely related to the prick test reactions. ( ...1978348345
mites in house dust in the stockholm area.today there is no doubt that mites (and especially species within the pyroglyphidae family: dermatophagoides pteronyssinus and dermatophagoides farinae) are an important agent in dust allergies. the author reports on a study of the occurrence of mites in house dust in the environment of allergic persons in the stockholm area. 201 private homes, 13 farmers' houses and two hospitals have been studied. in 12% of the homes several different types of mite were found in dust in relatively small number ...1979378014
house dust, mite (d. farinae) and cockroach allergy in a midwestern population.by using three different skin testing techniques and rast, this study explores the relationship among house dust, mite (dermatophagoides farinae) and cockroach allergy in atopic patients in the kansas city area. results suggest that the cockroach antigen may be as important as the mite in the etiology of house dust allergy in this population.1979426325

enzymatic hydrolysis of allergens extracted from the house dust mite, dermatophagoides farinae.the crude extract of the house dust mite, dermatophagoides farinae, was subjected to enzymatic hydrolysis by the use of trypsin and pronase. the hydrolysate was fractionated by gel-filtration and ion-exchange chromatographies. allergenic activities of the fractions were measured by prausnitz-küstner type skin test on guinea pig skin with homocytotropic antibodies. major allergenic component passed through anion and cation dowex resins but its yield was not large. on gel-filtration, the mite dige ...19751185896
population structure and dynamics of the house dust mite dermatophagoides farinae (acarina: pyroglyphidae) in czechoslovakia.the relationship between the room microclimate and population dynamics of the house dust mite dermatophagoides farinae hughes, 1961 was studied. a distinct increase of the mite population density in mattresses was observed only in the period, when the monthly means of relative air humidity in rooms were higher than 47-50%, denominated by the author as critical air humidity. this is a considerably lower value than that relevant to dermatophagoides pteronyssinus (trouessart, 1897) from the paper b ...19751193484
further studies in allergenic identity between house dust and the house dust mite, dermatophagoides farinae hughes, 1961.the sera from 99 japanese asthmatic subjects were studied for the allergenic similarity between house dust and the house-dust mite, dermatophagoides farinae hughes, 1961. a close correlation (correlation coefficient 0.92, p less than 0.01) was obtained between house dust and the mite with the radioallergosorbent test (rast). high correlation was not noted between house dust and other allergens such as smoky brown cockroach, periplaneta fuliginosa, aspergillus, book lice, liposcelis bostricophilu ...19751200424
[hyposensitization with extracts of house dust and mites adsorbed on aluminium-hydroxyde (allpyral) in children with bronchial asthma (author's transl)].since the year 1971 the authors have treated 39 asthmatic patients with allpyral house dust vaccins containing dermatophagoides farinae extract. 16 of these children previously had similar treatment, but without mite extract, and this was changed because of the poor results. patients allergic only to house dust and mites were selected according to the skin tests. skin reaction to mite extract did not change even in symptomfree children in contrast to the decrease or disappearance of skin sensiti ...19761264078
inhibition of mite protease (df-protease) with protease inhibitors.a protease from house dust mite(dermatophagoides farinae) having high specificity towards a substrate of blood coagulation factor xiia catalyzes the activation of kallikrein-kinin system in plasma (takahashi et al., 1990). to prevent the formation of kinin by the mite-protease, inhibition of the protease with its inhibitors was tested in vitro and in vivo. its kinetic studies revealed that ki values are 3.9 x 10(-10) m for aprotinin, 3.0 x 10(-9) m for soybean trypsin inhibitor (kunitz) and 2.5 ...19921282814
dust from carpeted and smooth floors. v. cat (fel d i) and mite (der p i and der f i) allergen levels in school dust. demonstration of the basophil histamine release induced by dust from classrooms.sixty dust samples from schools in norway were analysed for major allergens from cat and mite after sampling with the regularly used vacuum cleaners for 5 days and with a new model vacuum cleaner for 10 days, respectively. the major feline allergen felis domesticus allergen i (fel d i) was detected in all the classrooms, with ranges from 12 to 16,840 ng/m2 floor area. the mean fel d i concentration was about 11 times higher per unit area carpeted floors as compared with smooth floors after the 1 ...19921283110
[elevated production of gm-csf by peripheral blood mononuclear cells stimulated with mite antigens in children with bronchial asthma].granulocyte-macrophage colony-stimulating factor (gm-csf) is a cytokine which can stimulate the proliferation and functions of eosinophils and neutrophils. we have studied whether peripheral blood mononuclear cells (pbmc), obtained from children with stable asthma, can produce gm-csf through stimulation with crude dermatophagoides farinae (df) antigen in vitro. the patients had strongly positive rast results for house dust mite df. levels of gm-csf in pbmc culture supernatant were assayed by an ...19921290408
[clinical significance of igg4 antibody determination in children against egg white, milk, soybean and dermatophagoides farinae].the measurement of ige and igg4 antibodies against egg white, milk, soybean and dermatophagoides farinae was performed by fast (fluorescence allergosorbent test) using 21 serum samples obtained from non-allergic children and 160 serum samples from atopic children with bronchial asthma and/or atopic dermatitis. their antibody levels were evaluated for any association with disease severity and for clinical significance in establishing diagnosis. it was found that children with bronchial asthma sho ...19921290413

[allergen-stimulated release of prostaglandin e, f2 alpha by alveolar macrophages from patients with allergic asthma].bronchoalveolar lavage (bal) was performed in healthy subjects (n = 12) and patients with bronchial asthma (n = 11). between the two groups there was no significant difference in the total number of cells and the percentage of alveolar macrophages (am), lymphocytes (l), neutrophils (n) and eosinophils (e) in bal fluids. when am were cultured in vitro and stimulated with dermatophagoides farinae (dpf) antigen, the amount of pge released by am was significantly increased in the asthmatics. when th ...19921306779
[false positive reaction in measurement of allergen-specific ige--comparison of 3m ige fast-plus test using polystyrene well as adsorbent with phadezym rast].irrelevant ige binding to cellulose discs is known to give false positive results in phadezym rast (pharmacia) for the estimation of allergen-specific ige in serum. we investigated fast-plus test (3m diagnostic systems), an enzyme-linked sandwich type fluoro-allergo-sorbent test in which a particular allergen was coated to polystyrene well. phadezym rast and cap rast (pharmacia) using cellulose-derivative discs as adsorbent were used as reference methods. patients' sera which gave negative blank ...19921307629
cloning and expression of cdna coding for the major house dust mite allergen der f ii in escherichia coli.a cdna library corresponding to mite protein was screened using anti-der f ii, a major allergen from the house dust mite dermatophagoides farinae, antibody. three possible clones were obtained that contained cdna fragments coding for der f ii, and the nucleotide sequences of the fragments were determined. there were minor differences observed affecting the deduced amino acid sequence among the three cdna fragments. the amino acid sequence of the purified native der f ii protein could be analyzed ...19911368682
identification of allergenic epitopes on der f i, a major allergen of dermatophagoides farinae, using monoclonal antibodies.the antigenic and allergenic structure of der f i, a major allergen of the house dust mite dermatophagoides farinae (df) was investigated by means of a panel of 11 selected monoclonal antibodies (mab) obtained from balb/c mice immunized with purified der f i. the species specificity of these mab, tested with der f i and der p i--the homologous allergen from dermatophagoides pteronyssinus--was generally restricted to der f i since 10 out of 11 mab reacted only with this allergen. epitope specific ...19921371821
monitoring of mite dermatophagoides farinae allergen-specific igg and igg subclass distribution in patients on immunotherapy.house dust mite d. farinae and der f ii-specific igg and igg subclass responses were evaluated in 32 adults with perennial rhinitis undergoing immunotherapy for 1 year by means of igg-rast and elisa. the elisa method, which is based on subclass-specific monoclonal antibodies, could detect 0.5-1.5 ng/ml of specific antibodies. d. farinae and der f ii-specific igg4 antibodies increased continuously as immunotherapy proceeded, while total igg and igg1 antibodies reached a plateau value 6 months aft ...19921378700
[inhibitory activity of mite igg4, antibody on antigen-induced histamine release from human peripheral blood leukocytes].several reports have yielded conflicting results on the role of igg4 antibody on the surfaces of target cells in immediate type allergy. this study was performed to elucidate whether igg4 antibody inhibits ige-mediated histamine release from target cells after antigenic stimulation, and whether it has reaginic activity. serum was obtained from patients with nasal allergy receiving specific immunotherapy for housedust and mites. ige and igg4 were enriched affinity chromatographically using monocl ...19921380985
the allergen specific b-cell response during immunotherapy.the paper examines the allergen specific b-cell response in peripheral blood from patients undergoing immunotherapy with house dust mite extract. the 12 patients were part of a double blind placebo controlled study, and they were treated with either dermatophagoides pteronyssinus (n = 4), dermatophagoides farinae extract (n = 3) (alutard sq, alk, denmark) or placebo (n = 5). blood was taken every fortnight on day seven after hyposensitization and tested for igm, igg, iga and ige antibody secreti ...19921393764
[sensitization to house dust mites: importance and possibilities of allergen elimination].house-dust mite allergy (dermatophagoides pteronissinus and dermatophagoides farinae) is an important causative factor for allergic asthma or rhinitis in children. there is a clear relationship between the degree of allergen exposure and the subsequent development of asthma or the risk of sensitisation. it is therefore useful to know how intensive a patient is exposed to house-dust allergen. this can be managed either by counting all mites in the dust, by measuring of the content of der p i and ...19921434335
is a trypsin-like protease of mites a der f iii allergen?a trypsin-like protease was purified from mite (dermatophagoides farinae) fecal extract. in sds-page, the mite trypsin-like protease showed a single band at 34 kd. the purified trypsin-like protease possessed potent allergenic activity. both the twenty n-terminal amino acid sequence and the amino acid composition of the purified protease were very similar to those of der f iii. these data strongly suggest that the trypsin-like protease in the mite is a der f iii allergen.19921444822
[evaluation of oriton ige, a new kit for measurement of allergen specific ige antibodies].to determine whether oriton ige kit, a new kit for the measurement of allergen-specific ige antibodies, is useful in screening allergen-specific antibody, we measured the titers of ige antibodies against 11 different allergens (house dust 2, dermatophagoides farinae, japanese cedar, ragweed pollen, egg white, milk, cat epithelium, dog epithelium, candida, alternaria and aspergillus) with the oriton ige kit, and the results were compared to those of intradermal tests and rast in 103 allergic pati ...19921444828

cd4+ cd45ra+ t cells modulate allergen-induced interleukin 2 responsiveness in human lymphocytes.peripheral blood lymphocytes from nonallergic individuals acquired responsiveness to interleukin 2 (il2) after stimulation with ovalbumin (ova) or dermatophagoides farinae (df) antigens when they were pretreated with the cd45ra antibody, which has been shown to define the suppressor inducer subset of cd4+ cells and also to block its suppressor activity. the effect provided by the cd45ra antibody was lost if the lymphocytes had initially been activated with the ova of df antigens. the magnitude o ...19921531787
[studies on igg subclass antibodies in adult asthma. 2. changes in serum antigen specific igg subclass antibodies for aging and intractability in asthmatics].to clarify the factors which induce intractable asthma, the level of serum igg subclass antibodies to mite (dermatophagoides farinae) and candida antigens (candida albicans) for aging and severity was investigated in 230 bronchial asthmatics (male: 117, female: 113) aged 6-81 years old (mean age = 40). total ige level and ige antibodies to mite and candida antigens were measured by radioimmunosorbent test (rist) and radioallergosorbent test (rast), respectively. the serum level of igg and igg1 a ...19921554328
trimodality of serum dermatophagoides farinae-specific ige rast levels in atopic dermatitis patients.serum dermatophagoides farinae-specific ige rast (df ige) levels were assayed in 122 atopic dermatitis patients at the first examination. from the statistical study, we found trimodality of the individual variability of serum df ige levels. we hypothesize that serum df ige levels may be controlled by a pair of allelic genes.19921582702
[eosinophil colony stimulating factor production by lymphocytes from patients with bronchial asthma].to investigate the eosinophilia in patients with bronchial asthma (ba), we examined the release of eosinophil colony stimulating factor (eo-csf) from blood mononuclear cells (mnc) and lymphocytes from ba. we also investigated the effect of specific antigens on the release of eo-csf to determine its relation to other known eo-csfs. 1) phytohaemagglutinin (pha) and interleukin (il)-2 at optimal concentrations stimulated mononuclear cells from ba, and induced eo-csf release. in contrast, mnc from n ...19921625398
the house dust mite dermatophagoides pteronyssinus is the most important allergen on the island of mauritius.to determine the relative importance of mites as a cause of allergic sensitivity and asthma on the western indian ocean island of mauritius, we measured specific ige antibodies to common inhalant allergens in sera from mauritians claiming to have allergic symptoms and we examined house dust samples for evidence of mites and their allergens. seventy-two of the 110 sera tested (65%) contained detectable ige antibody to at least one mite, mould or pollen allergenic extract. by far the most prevalen ...19921628251
reduction of environmental mites improved atopic dermatitis patients with positive mite-patch tests.during a series of studies on the involvement of house dust mite antigens in 183 cases of atopic dermatitis, we observed an improvement in two patients following the removal of mites from their environment by means of a thorough housecleaning and replacement of the mattress. both patients manifested the typical clinical skin lesions of atopic dermatitis and had similar laboratory findings. although the serum ige concentrations and specific ige to dermatophagoides pteronyssinus and dermatophagoid ...19921640023
skin cross-reactivity between dermatophagoides pteronyssinus and storage mites in atopic children.in our environment, some children with allergic respiratory pathology do not have as favorable an evolution as expected after three years of immunotherapy with d. pteronyssinus extracts. we believe this is due to the existence of other mites whose antigenic determinants are not present in d. pteronyssinus. the purpose of the present study was to evaluate skin cross-reactivity between dermatophagoides and storage mites in children with bronchial asthma. fifty patients were chosen, with a mean age ...19911669575
ovalbumin-specific induction of il2 responsiveness in lymphocytes from patients with hen egg allergy and its regulation by the culture supernatant of normal lymphocytes.interleukin 2 (il2) responsiveness of ovalbumin (ova)-stimulated lymphocytes from patients with hen-egg allergy was studied. the number of viable cells of 5-day cultured lymphocytes stimulated with ova was increased by an additional three days incubation with recombinant il2. this phenomenon was not observed when the lymphocytes of patients allergic to ova but not to dermatophagoides farinae extract antigen (df) were stimulated with df. normal lymphocytes stimulated with ova expressed tac antige ...19901688691
basophil histamine release and airway response to mite allergen in atopic dermatitis.twelve patients with atopic dermatitis (ad) were subjected to in vitro histamine release from peripheral blood leukocytes (basophils) and in vivo bronchial inhalation challenge using house dust mite (dermatophagoides farinae) allergen. not only seven patients with asthmatic history but also five patients without asthma responded to both the in vitro and the in vivo challenges. a significant correlation was observed between hr30 (a mite concentration producing a 30% release of total cellular hist ...19901699458
[histamine release test of patients allergic to the house dust mite, dermatophagoides farinae--analysis according to various clinical backgrounds].since the histamine release test is more complicated than some of the other tests, there are smaller number of clinical studies employing this method. in this study, we selected 25 males and 20 females, allergic to the house dust mite, to undergo the histamine release test challenged by 7 serial concentrations of the mite allergen diluted from 10 times to 10 million times the original allergen at a concentration of 0.01%. histamine release was expressed as a percentage of the total histamine con ...19911711309
the distribution of cat and dust mite allergens on wall surfaces.this study was undertaken to determine the distribution of cat and dust mite allergens on wall surfaces and to assess the value of wall-wipe samples as a measure of allergen exposure. paired samples were collected from 31 homes, 20 homes with cats and 11 homes without, by vacuuming 1 m2 of carpet and by wiping 1 square foot of an adjacent wall. felis domesticus allergen i (fel d i) was detected in 30 of 31 settled dust samples (range, not detectable to 832,000 mu/gm; median, 10,250 mu/gm) and in ...19921730832

synthesis of biologically active recombinant der f ii.a cdna library corresponding to mite protein was screened employing anti-der f ii antibody. two possible clones containing plasmids pfl1 and pfl11 were obtained. the two plasmids had insertions of about 500 basepairs. the dna sequences of the two insertions were determined, from which the amino acid sequences were deduced. the amino acid sequence of the purified native der f ii protein could be determined to 45 residues from the n terminus. as a result of comparison, we concluded that the cdnas ...19911937898
preparation of specific monoclonal antibodies against der f ii one of the major allergens of dermatophagoides farinae.five monoclonal antibodies (mabs) against der f ii, one of the major allergens of dermatophagoides farinae, were obtained from mice immunized with the allergen. western blotting analysis revealed that only one mab, named 15e11, was specific for der f ii, whereas the other mabs crossreacted with der p ii, one of the major allergens of dermatophagoides pteronyssinus. these results indicate the possibility that the der f ii-specific mab, 15e11, is a useful reagent for specific determination of der ...19911958138
house dust mites in the west indies.the purpose of this study was to determine the role of the house dust mite. dermatophagoides pteronyssinus (dp) and dermatophagoides farinae (df), in allergic diseases on the island of st. lucia. dust samples were taken from living quarters of patients and hospital volunteers. the level of df antigen in these living quarters was measured by rast inhibition. all samples showed df antigen within a large range of values. prick tests were done to house dust mite and molds on 91 patients with suspect ...19912014932
sequence analysis of cdna coding for a major house dust mite allergen, der f i.a cdna clone coding for der f i, a major allergen from the house dust mite dermatophagoides farinae has been isolated and sequenced. it codes for a putative 18-residue signal peptide, an 80-residue proenzyme region, and a 223-residue mature protein with a derived molecular weight of 25,191. the deduced amino-acid sequence shows significant homology to other cysteine proteases in the proregion as well as in the mature protein. sequence alignment of the mature der f i protein with the homologous a ...19912021874
cdna encoding the major mite allergen der f ii.cdna encoding the major house dust mite allergen der f ii from dermatophagoides farinae was amplified using the polymerase chain reaction and cloned into e. coli. it encoded a 129-residue protein with a calculated molecular weight of 14,021 d and had the expected high homology (88%) with der p ii including the absence of n-glycosylation sites and conserved cysteine residues. these results are consistent with the high degree of antibody crossreactivity and may help identify the differences in t-c ...19912021876
dermatophagoides farinae allergen levels from two different sources within the same home: evaluation of two different collection techniques.the allergens produced by the dust mites of the dermatophagoides genus are now recognized as one of the major allergens in house dust. quantification of those allergens helps to evaluate the effectiveness of avoidance measures and specific treatments aimed at the reduction of the mites and their allergens. the present study was undertaken to compare the level of d. farinae allergen collected by a standard vacuum cleaner with a disposable bag and the level of allergen collected by a hand-held vac ...19912035907
triggering of the vascular permeability reaction by activation of the hageman factor-prekallikrein system by house dust mite proteinase.a 30-kilodalton (kda) proteinase from the house dust mite dermatophagoides farinae (df-proteinase) was recently purified (takahashi et al. (1990) int. arch. allergy appl. immunol. 91, 80-85). in this paper we detailed the biological activities of the df-proteinase. the activation of the kinin cascade by df-proteinase was examined in vitro by using purified guinea pig hageman factor (hf), prekallikrein (pk) and high-molecular-weight kininogen (hmwk) and the effect of this proteinase on endogenous ...19912043681
house-dust-mite allergens: a review.the house-dust mites, dermatophagoides farinae, d. pheronyssinus and euroglyphus maynei are cosmopolitan inhabitants of the homes of humans worldwide. these mites are the sources of multiple potent allergens that trigger allergic reactions in house-dust-mite-sensitive individuals. many laboratories using widely varied mite materials and allergic sera, and biochemical and immunological assays, have isolated and characterized, to varying degrees, some of the allergens produced by these mites. the ...19912044430
[eosinophil chemotactic activity in the supernatant of mononuclear cell culture stimulated with specific antigen].peripheral blood mononuclear cells (pbmc) separated from patients with asthma who were sensitive to dermatophagoides farinae (df) were cultured in alpha-medium for 5 days at 37 degrees c in 5% co2, in the presence or absence of 10 ng/ml of df antigen. eosinophils were purified from the peripheral blood of patients with eosinophilia who were not sensitive to df. eosinophil chemotactic activity (eca) was tested using a modified boyden chamber method. eca in the supernatant of pbmc stimulated with ...19912069510
[suppressive effect of saibokuto on dermatophagoides farinae (df) antigen-induced il2 responsiveness of lymphocytes from asthmatic children].the effect of saibokuto on induction of antigen specific il2 responsiveness was studied. saibokuto suppressed the induction of il2 responsiveness induced by dermatophagoides farinae (df) antigen in peripheral blood mononuclear cells from patients with bronchial asthma. saibokuto-pretreated adherent cells failed to present df antigen to non-adherent responding t cells for induction of their il2 responsiveness. the same dose of saibokuto had no impact on non-adherent cells. saibokuto also suppress ...19902073141
[comparison of the cutaneous response to dermatophagoides farinae from 2 separate laboratories].we compared in 50 asthmatic children, the cutaneous response by skin prick test to dermatophagoides farinae with antigens from two different laboratories (freeman and hollister). in order to evaluate the importance of the selection, conservation and standardization of the allergic extracts. 27/50 children had a similar response, in those cases (6) where freeman was doubtful and hollister positive the allergic etiology was confirmed by rast and challenge tests. in 5 cases, freeman was negative an ...19902089605
[anti-cd45r antibody-treated normal lymphocytes respond to interleukin 2 (il2) after stimulation by allergens].interleukin 2 (il2) responsiveness was specifically induced by dermatophagoides farinae (df) antigen in df-sensitized lymphocytes from asthmatic children, but not in normal lymphocytes. df-induced il2 responsiveness was also observed in normal lymphocytes pretreated (day 0) with anti-cd45r antibody, which recognize suppressor inducer subset among cd4+ t cells. however anti-cd45r antibody was no longer effective when the lymphocytes were cultured for more than one day with the antigen, suggesting ...19902140934
