value of sputum culture in diagnosis of pneumococcal pneumonia.in our laboratory, culture of sputum was extremely useful in diagnosing the etiology of pneumococcal pneumonia. of 31 consecutive patients with bacteremic pneumococcal pneumonia, 29 (94%) had streptococcus pneumoniae cultured from sputum. recovery of pneumococci in culture was enhanced by anaerobic incubation as well as by a plate bile test and an optochin disk on a primary blood agar plate.197718489
specificity of cytoplasmic and cell-wall antigens from four species of phytophthora.cytoplasmic and cell-wall antigens and antisera were prepared from four phytophthora species, and cell-wall antigens were prepared from two pythium species. immunodiffusion of the pythium and phytophthora cell-wall antigens showed that the two pythium species did not cross-react with the phytophthora cell-wall antisera. immunodiffusion analysis of both cell-wall and cytoplasmic antigens of phytophthora revealed some degree of specificity between species but not between a1 and a2 mating types in ...197658961

[two-layer rocket-electrophoresis on cellulose-acetate membranes].two-floor rocket-electrophoresis on gelatinated acetate cellulose membrane "cellogel" has been developed. the method is based on electroimmunodiffusion detection of the antigen on the acetate-cellulose membranes, containing monospecific antiserum of the test-system. the procedure is followed by the detection of precipitation bands ("rockets") by staining, if the reaction is conducted in the visible zone or by the further treatment of the acetate-cellulose strips, containing invisible precipitate ...197993004
the binding of streptomycin to ribonucleotides.incubation of streptomycin (sm) with [32p]5'-ribonucleotides at ph 7.0 produces fractions that migrate towards the cathode in high voltage electrophoresis (hve) separations at ph 3.5. sm appears to interact with pg, pa and pc but not with pu. the appearance of these [32p]-labeled fractions is dependent on incubation time and sm concentration. incubation of nucleotides with dihydrostreptomycin (dsm) or sm reduced with sodium cyanoborohydride (nabh3cn) at ph 5.0, does not produce detectable change ...197993596

reversal of serotonin vasodilatation in the dog external carotid bed by sympathetic denervation.in view of the conflicting reports of both constrictor and dilator effects of serotonin on the external carotid vascular bed of dogs, the influence of intraarterial infusions of the amine on blood flow through this territory was assessed by electromagnetic flowmeter techniques. in anesthetized intact dogs, serotonin produced dose-related increases in flow. these vasodilator responses were markedly diminished after ipsilateral vagotomy and were followed by delayed vasoconstriction, which then bec ...197994376
hemodynamic consequences of administration of phentolamine or nitroprusside with dopamine in the dog.dopamine was administered intravenously to two groups of anesthetized dogs at rates of 2.5, 5, 10 and 20 micrograms/kg/min. after the 20 micrograms/kg/min infusion, mean arterial pressure, heart rate, renal blood flow, mean pulmonary arterial pressure, and left ventricular filling pressure were increased. the dopamine infusion was continued in both groups of animals. in group i phentolamine was administered at rates of 2.5, 5, and 7.5 micrograms/kg/min; in group ii sodium nitroprusside at rates ...197994404

assessment of immunocytochemical techniques with particular reference to the mixed-aggregation immunocytochemical technique.antibody-mediated assays are believed to be the most sensitive and specific techniques for antigen localization available to date, but they need extensive controls if specific and quantitative results are to be obtained. problems arise from both the antibody source and the antigen in the tissue preparation. specificity of the antibody is often difficult to prove in any direct or indirect antibody technique. when markers are coupled to antibody, binding and diffusion properties of the antibody ar ...197995185
behaviour of solute and particle markers in the stomach of sheep given a concentrate diet.1. fistulated sheep given a concentrate diet were used to study the behaviour of solute ([51cr]edta) and particle ([103ru]phenanthroline) markers in the stomach under conditions of continuous feeding. 2. an injection of a mixed dose of [51cr]edta and [103ru]phenanthroline was given into the rumen and the time course of marker concentrations in the rumen and the abomasum was recorded. the curves were analysed on the assumption that the stomach of the sheep could be represented as two mixing compa ...197896850
the bilateral response of the temporal muscle in the rhesus monkey (macaca mulatta) to short-term detachment of the muscle and increased loading of the mandible.the electromyographic background activity of the temporal muscle was observed after short-term detachment of the muscle and with increased loads on the mandible. detachment of the muscle increased its mean spike discharge within the first 24 days but without significant changes in the spectral components. loading the mandible decreased both the spike discharge and spectral components.197798541

improved safety of splenoportography by plugging of the needle tract.splenoportography, a useful preoperative diagnostic procedure in patients with portal hypertension, has been largely discontinued because of the risk of hemorrhage. we have used both ivalon and gelfoam in particle and plug form to plug the needle tract after splenoportography. initial studies were conducted on six dogs. plugging of the needle tract by injection of suspensions of small gelfoam or ivalon particles resulted in intravasation of these particles into the splenic vein. this was demonst ...197898987
fibrinogen turnover in pregnant rabbits during the first and last thirds of gestation.although thrombin-mediated fibrinogen derivatives have been observed in normal pregnancy, an increased turnover of fibrinogen has been suspected but has not yet been demonstrated. fibrinogen turnover was studied in 14 pregnant and 10 nonpregnant female rabbits by use of purified rabbit 125i-labeled fibrinogen. in the last third of gestation the half life of fibrinogen was shortened by 45% and the fractional catabolic rate increased by 44% in comparison to the values for the first third of gestat ...197899036
interferon inducing activity of polyinosinic acid.although poly(i) is generally considered to be inactive as an interferon inducer, we have found several authentic poly(i) preparations to be effective inducers. their interferon inducing ability varied considerably from one cell system to another. in human diploid fibroblasts, primed with interferon and superinduced by cycloheximide and actinomycin d, all active poly(i) samples proved nearly as effective in inducing interferon as poly(i).poly(c). in primary rabbit kidney cell cultures, the activ ...197899486
electrophoretic strand separation of long dnas with poly (u,g) in agarose gels.we have found that binding of poly(u,g) to single-stranded dna decreases its mobility in 0.3% agarose gels. differential binding to the complementary strands of denatured duplex dna provides a simple method for strand separation. the method is shown to work with bacteriophage lambda dna, adenovirus dna and mtdna for tetrahymena pyriformis. in all cases the strand that binds more poly(u,g) in cscl gradients also binds more in gels. the separated strands can be directly blotted from the gel onto n ...197899728
[surgical and dietetic problems of subtotal resection of the small intestine in adults (author's transl)].the human body badly tolerates the extensive loss of the small intestine by resection. malabsorption of various substances is followed by severe deficiency symptoms. in recent years a longer survival with a satisfactory general condition was achieved by intensive postoperative care and adequate diet--in our own cases even in adults and in old age.1978101816

transcription and translation in a pleiotropic streptomycin-resistant mutant of escherichia coli.the role of the ribosomal protein s12 (streptomycin protein) in ribosome function and in other metabolic processes in the cell has been investigated. a spontaneous streptomycin-resistant strain of escherichia coli (sm3) carrying a mutation in the rpsl gene is deficient in its ability to induce the synthesis of the enzyme bets-galactosidase. it was demonstrated that the reduced rate of enzyme synthesis results from deficiencies in both the transcription of the lactose operon and translation of th ...1979104958
implications for adult roles from differential styles of mother-infant bonding: an ethological study.ethological observations of maternal and infant behaviors of nine vervet monkey pairs (cercopithecus aethiops sabaeus) showed the effects of differential styles of early maternal responsiveness on later infant competence. those infants receiving the least amount of maternal responsiveness and the most time-off the mother in the first 3 months of development were more socially competent at 6 months of age. the results are discussed within current ethological "attachment" theories. the detachment ...1979105083
halophilic, lactose-positive vibrio in a case of fatal septicemia.a halophilic vibrio species was isolated from blood cultures from a 59-year-old male with enteritis. the strain differed from vibrio parahaemolyticus and vibrio alginolyticus by its ability to ferment lactose, its production of beta-galactosidase, and its lower nacl tolerance. a report of this infection and a description of the isolate is presented.1979107189

a comparison between the free grafting of sliced and intact muscles in the rat-1.transverse slicing is a new technique whereby a mammalian muscle can be freely grafted with success. this method eliminates contamination of the early graft by surviving muscle fibres and allows one to measure the development of contractile properties on a uniform population of regenerating muscle fibres.?ioauthor1975124664
[influence of microflora on the metabolism of l-threonine at the level of the rat digestive tract].intestinal tissues are found to incorporate l threonine 14cu at faster rate in the absence of microflora. most of the radioactivity of the digestive contents occurs in the tca soluble compounds, except in the coecum and large intestine of the conventional rats where the microflora synthetized its own proteins from the labelled digestive material. free threonine is the main labelled soluble compound in the conventional rats digestive contents. in the other hand, in the germfree rats digestive con ...1976134804
adenosine 3':5'-cyclic monophosphate as mediator of catabolite repression in escherichia coli.measurements of intracellular adenosine 3':5'-cyclic monophosphate (camp) concentrations in e. coli under a variety of conditions show that levels of this nucleotide are well correlated with the rate of synthesis of beta-galactosidase (beta-d-galactoside galactohydrolase, ec 3.2.1.23) in both catabolite repression and transient repression. these results, combined with extensive genetic and in vitro studies from a number of laboratories on the role of camp in e. coli, provide strong support for t ...1975166384
cross-reactions of normal bovine serums to foot-and-mouth disease virus in plaque-reduction neutralization and radial immunodiffusion.serums from 150 cattle with no known exposure to foot-and-mouth disease (fmd) virus were tested by both the plaque-reduction neutralization (prn) technique and the radial immunodiffusion (rid) technique to evaluate the significance and the extent of cross-reactions in these tests. serums from 30 cattle from each of 5 locations were tested against representative viruses of each of the 7 types of fmd virus. high levels of cross-reactions with both the rid and prn techniques were found in serums of ...1975167625
immunological relationship between delta herpesvirus of patas monkeys and varicells-zoster virus of humans.the delta herpesvirus (dhv) which produced outbreaks of exanthematous disease in patas monkeys was shown to have a close immunological relationship to varicella-zoster (v-z) virus of man. immunization of rhesus monkeys with dhv or v-z virus resulted in the development of neutralizing antibodies to both viruses and also in the production of complement-fixing antibodies to v-z virus. immunoglobulin m neutralizing antibody to v-z virus was demonstrated in the serum of a rhesus monkey immunized with ...1975168152
an analysis of the mode of action of carbachol on the chick biventer cervicis nerve--muscle preparation.the mechanism of contracture evoked by carbachol in the isolated chick biventer cervicis nerve--muscle preparation was studied. at concentrations lower than 11 mum, carbachol progressively induced contracture at a rate much slower than did acetylcholine. a spontaneous increase of the response to carbachol, but not to acetylcholine, was observed 2-4 hr after isolation of the muscle. by contrast, no change of the response occurred in the denervated muscle. anticholinesterase treatment shifted the ...1976177299

plasma adrenocorticotropin levels in normal dogs.plasma concentrations of adrenocorticotropin (acth; corticotropin) were measured in 31 normal dogs (house pets) at rest. the mean concentration was 45.77 pg/ml of blood, with individual values ranging from 17 to 98 pg/ml of blood. the measurement of this hormone secreted by the anterior lobe of the pituitary gland (adenohypophysis) may aid in the diagnosis of spontaneous canine adrenal cortical disorders, both primary and secondary.1977201191
lysis of halophilic vibrio alginolyticus and vibrio costicolus induced by chaotropic anions.high concentration (1.0 m) of kscn, but not of nascn, induced lysis of slightly halophilic vibrio alginolyticus and moderately halophilic vibrio costicolus, and the decrease in absorbance of the cell suspension was complete after 30 min at 25 degrees c. replacement of k+ with na+ effectively prevented the lysis by scn-.k+ salts of no3-, br- and i-, however, induced no significant lysis. in electron micrographs, a prolonged exposure of the cells of v. alginolyticus to 1.0 m kscn displaced the nuc ...1977201300

effect of temperature, salts, ph, and other factors on the development of peritrichous flagella in vibrio alginolyticus.the development of peritrichous flagella and, consequently, swarming of vibrio alginolyticus depend on a complex relationship between temperature, salt concentrations and ph. at temperatures above 28 degrees c v. alginolyticus did not develop peritrichous flagella unless certain minimal concentrations of nacl are present: the higher the temperature, the higher the nacl concentrations required for peritrichous flagella synthesis. this requirement for nacl at high temperatures is much more pronoun ...1975242290

vibrio alginolyticus wound infection: case report and review.vibrio alginolyticus was associated with a littoral wound infection in an arizonan injured near the sea of cortez. while this is the first instance from this estuary, the relatively recent increase in the number of isolates suggests both that most coastal waters are a natural habitat of the organism and that probably many previous isolates were incorrectly identified in the past. mischaracterization most likely results from a gram-negative work-up routine deficient for the isolation of halophili ...1979378095
studies of yeast phenylalanine-accepting transfer ribonucleic acid backbone structure in solution by phosphorus-31 nuclear magnetic resonance spectroscopy.approximately 17 diester phosphates from the backbone structure of yeast trnaphe give rise to phosphorus resonances, which are resolved in its 31p nmr spectrum. to localize these diester phosphates within the trna structure, 31p nmr spectra of several chemically or enzymatically modified yeast trnaphe species were recorded. to this end selective modifications were performed in the anticodon, the dhu, and the t psi c loop. modifications, performed in different loop regions, give rise to perturbat ...1979383144
chemotactic responses of vibrio alginolyticus to algal extracellular products.a capillary assay was used to evaluate the chemotactic responses of vibrio alginolyticus to three common algal extracellular products. acrylate and glycolate attracted the motile marine bacterium. the peak response occurred with 10(-2) m of each chemical. acrylic and glycolic acid also attracted v. alginolyticus, with the peak response occurring at 5 x 10(-4) m of each chemical. higher concentrations of the organic acids resulted in a decreased response. the bacteria also displayed positive chem ...1979396019

mapping of antigenic sites to monoclonal antibodies on the primary structure of the f1-atpase beta subunit from escherichia coli: concealed amino-terminal region of the subunit in the f1.to analyze relationships between the ternary and primary structures of the beta subunit of escherichia coli f1 atpase, we prepared two monoclonal antibodies beta 12 and beta 31 against the beta peptide. these antibodies bind to the beta subunit but do not bind to the f1 atpase, resulting in no inhibition of the atpase activities. several different portions of the beta subunit peptide were prepared by constructing expression plasmids carrying the corresponding dna segment of the beta subunit gene ...19921373594
middle ear infection due to vibrio alginolyticus. bacteriological characterization.a 47-year-old male suffering from chronic purulent ear discharge was investigated bacteriologically in autumn 1977. the isolated strain was identified as vibrio alginolyticus. the fact that the patient spent his vacation at a black-sea resort every year strongly pleads for a direct inoculation of the halophilic vibrio.1979484269
characterization of a vibrio cholerae virulence factor homologous to the family of tonb-dependent proteins.irga is an iron-regulated virulence factor for infection in an animal model with classical vibrio cholerae strain 0395. we detected gene sequences hybridizing to irga at high stringency in clinical isolates in addition to 0395, including another classical strain of v. cholerae, three v. cholerae strains of the el tor biotype, three non-o1 isolates of v. cholerae, and individual isolates of vibrio parahaemolyticus, vibrio fluvialis, and vibrio alginolyticus. no hybridization to irga was seen with ...19921406279
prediction of transmembrane topology of f0 proteins from escherichia coli f1f0 atp synthase using variational and hydrophobic moment analyses.the a subunit, a membrane protein from the e. coli f1f0 atp synthase has been examined by fourier analysis of hydrophobicity and of amino-acid residue variation. the amino-acid sequences of homologous subunits from vibrio alginolyticus, saccharomyces cerevisiae, neurospora crassa, aspergillus nidulans, schizosaccharomyces pombe and candida parapsilosis were used in the variability analysis. by fourier analysis of sequence variation, two transmembrane helices are predicted to have one face in con ...19921445940

[membrane atpase of vibrio alginolyticus. ion transport activity and homology with f0f1-atpase from e. coli].f0f1-atpase has been isolated from the marine alkali-resistant bacterium vibrio alginolyticus. the enzyme subunits cross-reacted with antibodies against subunits alpha, beta, gamma, epsilon, and b of e. coli atpase. the purified atpase was reconstituted into liposomes effecting an atp-dependent uptake of h+. proton transport was inhibited by the atpase blockers dccd, triphenyltin, and venturicidin. na+ ions had no effect on atp-dependent proton transport. no atp-dependent transport of na+ was de ...19921457596
amiloride at ph 7.0 inhibits the na(+)-driven flagellar motors of vibrio alginolyticus but allows cell growth.amiloride, a specific inhibitor for the na(+)-driven flagellar motors of alkalophilic bacillus, is known to inhibit secondarily the growth of alkalophiles. the motility of a marine vibrio, v. alginolyticus, was almost completely inhibited by 2 mm amiloride either at ph 7.0 or 8.5. we found that this concentration of amiloride inhibited the cell growth completely at ph 8.5 but only slightly at ph 7.0. kinetic analysis of the inhibition of motility by amiloride at ph 7.0 showed that the inhibition ...19921459240

vibrio alginolyticus ("achromobacter") collagenase: biosynthesis, function and application.bacterial collagenase from aerobic non-pathogenic vibrio alginolyticus chemovar iophagus ("achromobacter" collagenase, ec 3.4.24.08) is an inducible extracellular metallo-proteinase. production of vibrio collagenase is induced specifically by collagen or by its macromolecular fragments. on the cell surface is expressed a specific receptor recognizing collagen structure. the study of natural inducers led to synthetic peptides with inducing properties. vibrio collagenase cleaves collagen helical c ...19921480012
gel filtration of sialoglycoproteins.the role of sialic acid in the gel-filtration behaviour of sialoglycoproteins was investigated by using the separated isoenzymes of purified human liver alpha-l-fucosidase and several other well-known sialic acid-containing glycoproteins (fetuin, alpha1-acid glycoprotein, thyroglobulin and bovine submaxillary mucin). for each glycoprotein studied, gel filtration of its desialylated derivative gave an apparent molecular weights much less than that expected just from removal of sialic acid. for th ...1978356853
purification and characterization of l-2,4-diaminobutyrate decarboxylase from acinetobacter calcoaceticus.acinetobacter calcoaceticus atcc 23055 produces a large amount of 1,3-diaminopropane under normal growth conditions. the enzyme responsible, l-2,4-diaminobutyrate (daba) decarboxylase (ec 4.1.1.-), was purified to electrophoretic homogeneity from this bacterium. the native enzyme had an m(r) of approximately 108,000, with a pi of 5.0, and was a dimer composed of identical or nearly identical subunits with apparent m(r) 53,000. the enzyme showed hyperbolic kinetics with a km of 1.59 mm for daba a ...19921512577
isolation of vibrio alginolyticus from wounds and blood of a burn patient.isolation of vibrio alginolyticus from the wounds and blood of a burn patient is reported. numerous tissue biopsy sites as well as one blood culture yielded the organism. the case history and a review of the hospital course of the patient is provided where relevant to the presence and identification of the organism. the bacteriologic, biochemical, and anti-microbial identification of the organism is described. taxonomy, habitat, and pathogenicity of v. alginolyticus are discussed.1977335885
properties of respiratory chain-linked na(+)-independent nadh-quinone reductase in a marine vibrio alginolyticus.the respiratory chain of a marine vibrio alginolyticus contains two types of nadh-quinone reductase (nqr): one is an na(+)-dependent nqr functioning as an na+ pump (nqr-1) and the other is an na(+)-independent nqr (nqr-2). nqr-2 was purified about 55-fold from the membrane of mutant nap-1 which is devoid of nqr-1, and its properties were compared with those of nqr-1. in contrast to nqr-1, the purified nqr-2 does not require any salts for activity and is not inhibited by up to 0.4 m salts. the op ...19921543699

the highest gene concentrations in the human genome are in telomeric bands of metaphase chromosomes.chromosome in situ suppression hybridization has been carried out on human metaphase chromosomes to localize the g+c-richest human dna fraction (which only represents 3.5% of the genome), as isolated by preparative equilibrium centrifugation in cs2so4/3,6-bis(acetatomercurimethyl)-1,4-dioxane density gradient. this fraction essentially corresponds to isochore family h3. the rationale for carrying out this experiment is that this isochore family has, by far, the highest gene concentration, the hi ...19921594593
characteristic differences in the mode of quinone reduction and stability between energy-coupled and -uncoupled nadh-quinone reductases from bacterial respiratory chain.bacterial respiratory chain has two types of nadh-quinone reductase (nqr): one is energy-coupled (type-1) and the other had no energy-transducing capacity, that is, energy-uncoupled (type-2). each of the nadh-reacting flavoprotein subunits of nqr-1 from escherichia coli and the marine vibrio alginolyticus reduced quinone to semiquinone radicals by the one-electron transfer pathway and was very sensitive to preincubation with nadh. on the other hand, the nqr-2 from these bacteria reduced quinone ...19921628743
