effect of solvent phase transitions on enzymatic activity and structure of laccase from coriolus hirsutus.the effect of solvent phase transitions on catalytic activity and structure of the active site of laccase produced by the basidiomycetes coriolus hirsutus 072 was studied. as shown by small-angle x-ray scattering, laccase exists in solution as a mixture of monomeric and aggregated particles in the percent ratio 85:15. this ratio did not change on phase transitions. a complex nature of laccase activity dynamics during thawing and further heating to 20 degrees c was shown. spontaneous oxidation of ...200919463091
structure of native laccase from trametes hirsuta at 1.8 a resolution.this paper describes the structural analysis of the native form of laccase from trametes hirsuta at 1.8 a resolution. this structure provides a basis for the elucidation of the mechanism of catalytic action of these ubiquitous proteins. the 1.8 a resolution native structure provided a good level of structural detail compared with many previously reported laccase structures. a brief comparison with the active sites of other laccases is given.200919465775
enhanced saccharification of alkali-treated rice straw by cellulase from trametes hirsuta and statistical optimization of hydrolysis conditions by rsm.a white rot fungus, identified as trametes hirsuta based on morphological and phylogenetic analysis, was found to contain efficient cellulose degrading enzymes. the strain showed maximum endoglucanase (eg), cellobiohydrolase (cbh) and beta-glucosidase (bgl) activities of 55, 0.28 and 5.0 u/mg-protein, respectively. rice straw was found to be a potentially good substrate for growth of t. hirsuta for cellulase production. statistical experimental design was used to optimize hydrolysis parameters s ...200919540109
effects of tyrosinase and laccase on oat proteins and quality parameters of gluten-free oat breads.effects of a trichoderma reesei tyrosinase (tyr) and a trametes hirsuta laccase (lac) on breadmaking performance of gluten-free oat flour were investigated by sds-page analysis of oat protein fractions, large deformation rheology, and microscopy of the doughs, as well as on the basis of specific volume and firmness of the gluten-free breads. tyr induced the formation of higher molecular weight proteins in the sds-page assay. microscopical analysis showed more intensive protein aggregation in the ...201121732697
pretreatment of paddy straw with trametes hirsuta for improved enzymatic saccharification.delignification of paddy straw with the white-rot fungus, trametes hirsuta under solid state fermentation, for enhanced sugar recovery by enzymatic saccharification was studied. t. hirsuta mtcc136 showed high ligninase and low cellulase activities. solid state fermentation of paddy straw with t. hirsuta enhanced carbohydrate content by 11.1% within 10days of incubation. alkali extracts of trametes pretreated paddy straw showed high absorbance at 205nm indicating high lignin break down. the amoun ...201222093976
Cloning of novel cellulases from cellulolytic fungi: Heterologous expression of a family 5 glycoside hydrolase from Trametes versicolor in Pichia pastoris.Total cDNA isolated from cellulolytic fungi cultured in cellulose was examined for the presence of sequences encoding for endoglucanases. Novel sequences encoding for glycoside hydrolases (GHs) were identified in Fusarium oxysporum, Ganoderma applanatum and Trametes versicolor. The cDNA encoding for partial sequences of GH family 61 cellulases from F. oxysporum and G. applanatum shares 58 and 68% identity with endoglucanases from Glomerella graminicola and Laccaria bicolor, respectively. A new G ...201122142722
oxidation of glycerol by 2,2,6,6-tetramethylpiperidine-n-oxyl (tempo) in the presence of laccase.glycerol has the potential of being a low-cost and extremely versatile building block. however, current transformation strategies such based on noble-metal-catalysts show several disadvantages including catalyst deactivation or negative environmental impacts. in this study glycerol was oxidized by 2,2,6,6-tetramethylpiperidine-n-oxyl (tempo) in the presence of laccase from trametes hirsuta. analysis of the reaction production indicated sequential oxidation to glyceraldehyde, glyceric acid and ta ...200919464170
direct electron transfer of trametes hirsuta laccase adsorbed at unmodified nanoporous gold electrodes.the enzyme trametes hirsuta laccase undergoes direct electron transfer at unmodified nanoporous gold electrodes, displaying a current density of 28μa/cm(2). the response indicates that thlc was immobilised at the surface of the nanopores in a manner which promoted direct electron transfer, in contrast to the absence of a response at unmodified polycrystalline gold electrodes. the bioelectrocatalytic activity of thlc modified nanoporous gold electrodes was strongly dependent on the presence of ha ...201223274541
a comparison of redox polymer and enzyme co-immobilization on carbon electrodes to provide membrane-less glucose/o2 enzymatic fuel cells with improved power output and stability.glassy carbon and graphite electrodes modified with films of enzyme and osmium redox polymer, cross linked with poly (ethylene glycol) diglycidyl ether, were used for elaboration of a glucose/o(2) enzymatic fuel cell. the redox polymers [os(4,4'-dimethoxy-2,2'-bipyridine)(2)(polyvinylimidazole)(10)cl](+) and [os(4,4'-dichloro-2,2'-bipyridine)(2)(polyvinylimidazole)(10)cl](+) were selected to facilitate transfer of electrons from the glucose oxidase (gox) active site to the t1 cu site of multicop ...201122005596
Isolation and evaluation of terrestrial fungi with algicidal ability from Zijin Mountain, Nanjing, China.Approximately 60 fungal isolates from Zijin Mountain (Nanjing, China) were screened to determine their algicidal ability. The results show that 8 fungi belonging to Ascomycota and 5 belonging to Basidiomycota have algicidal ability. Of these fungi, Irpex lacteus T2b, Trametes hirsuta T24, Trametes versicolor F21a, and Bjerkandera adusta T1 showed strong algicidal ability. The order of fungal chlorophyll-a removal efficiency was as follows: T. versicolor F21a > I. lacteus T2b > B. adusta T1 > T. ...201121887638
a new synthesis route for os-complex modified redox polymers for potential biofuel cell applications.a new synthesis route for os-complex modified redox polymers was developed. instead of ligand exchange reactions for coordinative binding of suitable precursor os-complexes at the polymer, os-complexes already exhibiting the final ligand shell containing a suitable functional group were bound to the polymer via an epoxide opening reaction. by separation of the polymer synthesis from the ligand exchange reaction at the os-complex, the modification of the same polymer backbone with different os-co ...201122209452
direct ethanol production from starch, wheat bran and rice straw by the white rot fungus trametes hirsuta.the white rot fungus trametes hirsuta produced ethanol from a variety of hexoses: glucose, mannose, cellobiose and maltose, with yields of 0.49, 0.48, 0.47 and 0.47 g/g of ethanol per sugar utilized, respectively. in addition, this fungus showed relatively favorable xylose consumption and ethanol production with a yield of 0.44 g/g. t. hirsuta was capable of directly fermenting starch, wheat bran and rice straw to ethanol without acid or enzymatic hydrolysis. maximum ethanol concentrations of 9. ...201122112911
a mediated glucose/oxygen enzymatic fuel cell based on printed carbon inks containing aldose dehydrogenase and laccase as anode and cathode.enzyme electrodes show great potential for many applications, as biosensors and more recently as anodes and cathodes in biocatalytic fuel cells for power generation. enzymes have advantages over metal catalysts, as they provide high specificity and reaction rates, while operating under mild conditions. here we report on studies related to development of mass-producible, completely enzymatic printed glucose/oxygen biofuel cells. the cells are based on filter paper coated with conducting carbon in ...201122305173
