rilyd4, a human cd59 inhibitor, enhances complement-dependent cytotoxicity of ofatumumab against rituximab-resistant b-cell lymphoma cells and chronic lymphocytic leukemia.ofatumumab is an anti-cd20 antibody recently approved for treatment of fludarabine and alemtuzumab refractory chronic lymphocytic leukemia (cll); it mediates much stronger complement-dependent cytotoxicity (cdc) than rituximab. human cd59, a key membrane complement regulator that inhibits cdc, is highly expressed in b-cell malignancies and its upregulation is an important determinant of the sensitivity of b-cell malignancies to rituximab treatment. previously, we have shown that the potent cd59 ...201121918174
A lingual abscess caused by Streptococcus intermedius.Lingual abscesses are rare. We describe a case in a healthy female with no recent history of trauma. The organism recovered by culture of drainage material collected prior to antibiotic treatment was Streptococcus intermedius, an organism recognized as flora of the oropharynx and associated with abscess formation. The isolate was resistant to clindamycin, which was the antibiotic therapy that the patient received.201122116986
role of streptococcus intermedius dnak chaperone system in stress tolerance and pathogenicity.streptococcus intermedius is a facultatively anaerobic, opportunistic pathogen that causes purulent infections and abscess formation. the dnak chaperone system has been characterized in several pathogenic bacteria and seems to have important functions in stress resistance and pathogenicity. however, the role of dnak in s. intermedius remains unclear. therefore, we constructed a dnak knockout mutant that exhibited slow growth, thermosensitivity, accumulation of groel in the cell, and reduced cyto ...201121822788
[brain abscess as the first clinical manifestation of isolated pulmonary arteriovenous malformation without rendu-osler disease].brain abscesses occur in 5 to 13 % of patients with pulmonary arteriovenous malformation (pavm), more often present in rendu-osler-weber disease or hereditary hemorrhagic telangiectasia (hht).201120466398

clustering of subgingival microbial species in adolescents with periodontitis.it has become increasingly recognized that groups of microorganisms interact within the subgingival plaque of adult subjects with periodontitis. it is much less clear, however, whether the consortia of microorganisms associated with periodontitis are different in early and more advanced cases of periodontitis. to investigate this point further, subgingival plaque was collected from six sites in 87 adolescents with periodontitis and 73 controls and the samples were analyzed for the detection of 1 ...201121410554
spiramycin resistance in human periodontitis microbiota.purpose: the occurrence of in vitro resistance to therapeutic concentrations of spiramycin, amoxicillin, and metronidazole was determined for putative periodontal pathogens isolated in the united states. materials and methods: subgingival plaque specimens from 37 consecutive adults with untreated severe periodontitis were anaerobically cultured, and isolated putative periodontal pathogens were identified to a species level. in vitro resistance to spiramycin at 4 μg/ml, amoxicillin at 8 μg/ml, an ...201121524712
human cd59 inhibitor sensitizes rituximab-resistant lymphoma cells to complement-mediated cytolysis.rituximab efficacy in cancer therapy depends in part on induction of complement-dependent cytotoxicity (cdc). human cd59 (hcd59) is a key complement regulatory protein that restricts the formation of the membrane attack complex, thereby inhibiting induction of cdc. hcd59 is highly expressed in b-cell non-hodgkin's lymphoma (nhl), and upregulation of hcd59 is an important determinant of the sensitivity of nhl cells to rituximab treatment. here, we report that the potent hcd59 inhibitor rilyd4 enh ...201121252115
application of a novel inhibitor of human cd59 for the enhancement of complement-dependent cytolysis on cancer cells.many monoclonal antibodies (mabs) have been extensively used in the clinic, such as rituximab to treat lymphoma. however, resistance and non-responsiveness to mab treatment have been challenging for this line of therapy. complement is one of the main mediators of antibody-based cancer therapy via the complement-dependent cytolysis (cdc) effect. cd59 plays a critical role in resistance to mabs through the cdc effect. in this paper, we attempted to investigate whether the novel cd59 inhibitor, rec ...201121258360
induction of experimental endocarditis by continuous low-grade bacteremia mimicking spontaneous bacteremia in humans.transient high-grade bacteremia following invasive procedures carries a risk of infective endocarditis (ie). this is supported by experimental endocarditis. on the other hand, case-control studies showed that ie could be caused by cumulative exposure to low-grade bacteremia occurring during daily activities. however, no experimental demonstration of this latter possibility exists. this study investigated the infectivity in animals of continuous low-grade bacteremia compared to that of brief high ...201121321073
mapping the intermedilysin-human cd59 receptor interface reveals a deep correspondence with the binding site on cd59 for complement binding proteins c8{alpha} and c9.cd59 is a glycosylphosphatidylinositol-anchored protein that inhibits the assembly of the terminal complement membrane attack complex (mac) pore, whereas streptococcus intermedius intermedilysin (ily), a pore forming cholesterol-dependent cytolysin (cdc), specifically binds to human cd59 (hcd59) to initiate the formation of its pore. the identification of the residues of ily and hcd59 that form their binding interface revealed a remarkably deep correspondence between the hcd59 binding site for i ...201121507937

Extracellular DNA in biofilms.Extracellular DNA (eDNA) is an important biofilm component that was recently discovered. Its presence has been initially observed in biofilms of Pseudomonas aeruginosa, Streptococcus intermedius, Streptococcus mutans, then Enterococcus faecalis and staphylococci. Autolysis is the common mechanism by which eDNA is released. In P. aeruginosa eDNA is generated by lysis of a bacterial subpopulation, under control of quorum sensing system. In E. faecalis autolysis proceeds in a fratricide mode, resul ...201122094562

polymicrobial subdural empyema: involvement of streptococcus pneumoniae revealed by lyta pcr and antigen detection.the authors report a case of a subdural empyema (sde) caused by a coinfection with streptococcus intermedius and streptococcus pneumoniae, initially considered a s. intermedius infection only. an otherwise healthy 11-year-old female was admitted to the hospital after 5 days of illness. symptoms were consistent with classical sde symptoms and progressed rapidly with finally somnolence before the first neurosurgical procedure despite relevant antibiotic treatment. primary mri showed an interhemisp ...201122707602
invasion of human aortic endothelial cells by oral viridans group streptococci and induction of inflammatory cytokine production.oral viridans group streptococci are the major commensal bacteria of the supragingival oral biofilm and have been detected in human atheromatous plaque. atherosclerosis involves an ongoing inflammatory response, reportedly involving chronic infection caused by multiple pathogens. the aim of this study was to examine the invasion of human aortic endothelial cells (haecs) by oral viridans group streptococci and the subsequent cytokine production by viable invaded haecs. the invasion of haecs by ba ...201021214874
sialidase of streptococcus intermedius: a putative virulence factor modifying sugar chains.a sialidase gene of streptococcus intermedius was cloned. it was most similar to nana, a major sialidase gene in streptococcus pneumoniae, and was expressed in escherichia coli. since the gene-knockout s. intermedius strain lost detectable sialidase activity, the gene might code, either solely or mainly, the glycosidase in the bacterial genome. polymerase chain reaction using the primers for the nana homologue in s. intermedius (described as nana below) showed that this sialidase gene was common ...201021118296
osteoradionecrosis contains a wide variety of cultivable and non-cultivable bacteria.direct microscopy, anaerobic culture and dna-dna hybridization have previously demonstrated an association between microorganisms and osteoradionecrosis (orn). the purpose of our study was to use culture independent molecular techniques to detect bacteria in necrotic bone lesions of the mandible after radiation therapy.201021523230
a high-affinity inhibitor of human cd59 enhances complement-mediated virolysis of hiv-1: implications for treatment of hiv-1/aids.many pathogenic enveloped viruses, including hiv-1, escape complement-mediated virolysis by incorporating host cell regulators of complement activation into their own viral envelope. the presence of complement regulators including cd59 on the external surface of the viral envelope confers resistance to complement-mediated virolysis, which may explain why human pathogenic viruses such as hiv-1 are not neutralized by complement in human fluids, even in the presence of high ab titers against the vi ...201019955519
characterization of streptococcus milleri group isolates from expectorated sputum of adult patients with cystic fibrosis.with the recent insights into the streptococcus milleri group (smg) as pulmonary pathogens in patients with cystic fibrosis (cf), we sought to characterize 128 isolates from the sputum of adults with cf, along with 45 isolates from patients with invasive diseases for comparison. the tests performed included lancefield grouping; tests for hemolysis; tests for the production of hyaluronidase, chondroitin sulfatase, dnase, proteases, and hydrogen peroxide; and pcr for the detection of the intermedi ...201020007382
diffusion-weighted mr imaging of intramedullary spinal cord abscess.early diagnosis and prompt initiation of adequate treatment are essential for clinical outcome in isca. we report a case in which dwi provided a more specific diagnosis than conventional mr imaging and allowed differentiation of a ring-enhancing lesion from intramedullary tumor. diagnosis was proved by pcr from csf (streptococcus intermedius). adequate antibiotic treatment was immediately initiated, and the patient recovered completely.201020019101
amplification of minute amounts of oral bacterial dna for real-time quantitative pcr analysis.high-throughput technologies for typing caries or health-associated bacterial populations including pcr, dna microarrays and next-generation sequencing techniques require significant amounts of bacterial dna. in clinical settings, the amount of sampled dna is often limited and amplification is therefore essential. protocols should be able to reproducibly amplify sequences in order to maintain initial sequence ratios and should not bias the representation of particular dna sequence types.201020881394
inerolysin, a cholesterol-dependent cytolysin produced by lactobacillus iners.lactobacillus iners is a common constituent of the human vaginal microbiota. this species was only recently characterized due to its fastidious growth requirements and has been hypothesized to play a role in the pathogenesis of bacterial vaginosis. here we present the identification and molecular characterization of a protein toxin produced by l. iners. the l. iners genome encodes an open reading frame with significant primary sequence similarity to intermedilysin (ily; 69.2% similarity) and vag ...201021169489
crystal structure of cytotoxin protein suilysin from streptococcus suis.cholesterol-dependent cytolysins (cdc) are pore forming toxins. a prototype of the cdc family members is perfringolysin o (pfo), which directly binds to the cell membrane enriched in cholesterol, causing cell lysis. however, an exception of this general observation is intermedilysin (ily) of streptococcus intermedius, which requires human cd59 as a receptor in addition to cholesterol for its hemolytic activity. a possible explanation of this functional difference is the conformational variation ...201021204001
acute subepicardial infarction associated with severe septic shock--insight in myocardial perfusion.isolated infarctions of the subepicardial myocardium without changes in subendocardium are extremely rare. we present an autoptic case with an acute subepicardial infarction of the right- and left-ventricular myocardium. a 53-year-old male was admitted to hospital with acute upper abdominal pain. clinical examination revealed an acute infero-lateral myocardial infarction. the patient succumbed to acute heart failure a few hours later. autopsy revealed numerous pulmonary abscesses due to suppurat ...201019945802

characterization of a streptococcal cholesterol-dependent cytolysin with a lewis y and b specific lectin domain.the cholesterol-dependent cytolysins (cdcs) are a large family of pore-forming toxins that often exhibit distinct structural changes that modify their pore-forming activity. a soluble platelet aggregation factor from streptococcus mitis (sm-hpaf) was characterized and shown to be a functional cdc with an amino-terminal fucose-binding lectin domain. sm-hpaf, or lectinolysin (lly) as renamed herein, is most closely related to cdcs from streptococcus intermedius (ily) and streptococcus pneumoniae ( ...200818553932
streptococcus cristatus attenuates fusobacterium nucleatum-induced interleukin-8 expression in oral epithelial cells.oral epithelial cells may be invaded by a polymicrobial intracellular flora, including pathogens together with commensals. various oral pathogens can induce the production of interleukin-8, a potent neutrophil chemotractant, in oral epithelial cells. evidence from the gut suggests that commensal species may modulate inflammatory responses to pathogens. the aim of this study was to examine the interleukin-8 responses of oral epithelial cells to an oral pro-inflammatory species, fusobacterium nucl ...200818942189
biofilm formation and autoinducer-2 signaling in streptococcus intermedius: role of thermal and ph factors.bacteria sense their population density using autoinducer (ai) signaling systems. the ai-2 signal is thought to mediate communication among and within bacterial species. streptococcus intermedius is a commensal organism frequently associated with periodontitis and purulent infections. we investigated the role of ai-2 signaling in s. intermedius biofilm formation under temperatures and ph levels relevant to human physiology.200818954356
streptococcus intermedius causing infective endocarditis and abscesses: a report of three cases and review of the literature.streptococcus intermedius is a member of the streptococcus anginosus group. clinical disease with s. intermedius is characterized by abscess formation and rarely endocarditis. identification of streptococcus intermedius is difficult, leading to the development of molecular methods to more accurately identify and characterize this organism.200818992173
streptococcus mutans and streptococcus intermedius adhesion to fibronectin films are oppositely influenced by ionic strength.bacterial adhesion to protein-coated surfaces is mediated by an interplay of specific and nonspecific interactions. although nonspecific interactions are ubiquitously present, little is known about the physicochemical mechanisms of specific interactions. the aim of this paper is to determine the influence of ionic strength on the adhesion of two streptococcal strains to fibronectin films. streptococcus mutans lt11 and streptococcus intermedius nctc11324 both possess antigen i/ii with the ability ...200818729493
heterologous expression ofa histone-like protein from streptococcus intermedius in escherichia coli alters the nucleoid structure and inhibits the growth of e. coli.escherichia coli failed to survive after transformation with a streptococcus intermedius histone-like protein gene (si-hlp) and its promoter-harbored plasmid. the promoter function of si-hlp in e. coli was determined using enhanced green fluorescence protein (egfp) gene as a reporter. the inhibitory effect of si-hlp on e. coli viability was verified by a tetracycline-inducible gene expression system. further study suggested that si-hlp may alter the bacterial nucleoid structure, leading to the g ...200818761672
identification and molecular analysis of betac-s lyase producing hydrogen sulfide in streptococcus intermedius.hydrogen sulfide (h(2)s) is a toxic gas that induces the modification and release of haemoglobin in erythrocytes; however, it also functions in methionine biosynthesis in bacteria. betac-s lyase, encoded by the lcd gene, is responsible for bacterial h(2)s production through the cleavage of l-cysteine. in this study, 26 of 29 crude extracts from reference and clinical strains of streptococcus intermedius produced h(2)s from l-cysteine. the capacities in those strains were not higher than those in ...200818927421
[when genetics matters: an unusual case of left-sided hemiparesis].we report the case of a 35-year-old woman who has been admitted to our emergency room because of the sudden onset of a left-sided hemiparesis. the physical examination showed disseminated teleangiectases on the upper and lower lip, on the mucosal surface of the tongue and on the skin. the cerebral ct scan presented a right-sided fronto-parietal lesion, more likely an abscess, confirmed on surgical removal. a meticulous family history showed the presence of similar clinical features among several ...200819016425

acetaldehyde production from ethanol by oral streptococci.alcohol is a well documented risk factor for upper digestive tract cancers. it has been shown that acetaldehyde, the first metabolite of ethanol is carcinogenic. the role of microbes in the production of acetaldehyde to the oral cavity has previously been described in several studies. in the present study, the aim was to investigate the capability of viridans group streptococci of normal oral flora to produce acetaldehyde in vitro during ethanol incubation. furthermore, the aim was to measure th ...200716859955
structural elements of the cholesterol-dependent cytolysins that are responsible for their cholesterol-sensitive membrane interactions.the pore-forming mechanism of the cholesterol-dependent cytolysins (cdcs) exhibits an absolute requirement for membrane cholesterol. the structural elements of the cdcs that mediate this interaction are not well understood. three short hydrophobic loops (l1-l3) and a highly conserved undecapeptide sequence at the tip of domain 4 of the cdc structure are known to anchor the cdc to the membrane. it has been thought that the undecapeptide directly mediates the interaction of the cdcs with a cholest ...200718077338
splenic abscess in an infant caused by streptococcus intermedius.we report a 20-month-old girl with splenic abscess. the patient was admitted to our hospital because of persistent high fever and abdominal pain. laboratory data showed leucocytosis and elevated c-reactive protein levels. abdominal computed tomography showed multiple low-density lesions in the spleen. these findings were consistent with a diagnosis of splenic abscess. she was successfully treated with ultrasonographically guided percutaneous drainage for 11 days and intravenous antibiotic for 17 ...200718095094
lemierre syndrome associated with dental infections. report of one case and review of the literature.the first publication on lemierre syndrome appears in 1936 by lemierre. it is defined as an "oropharynx bacterial infection characterized by the thrombophlebitis in the internal jugular vein, derived in a systemic septic embolism". in 81% of the cases, the fusobacterium necrophorum is the most frequent etiologic agent. fever is the most common symptom, but it can depending on the primary infection, tonsillitis, mastoiditis or odontogenic infection. according to the literature the mortality is ve ...200717767106
specific protein-membrane contacts are required for prepore and pore assembly by a cholesterol-dependent cytolysin.three short hydrophobic loops and a conserved undecapeptide at the tip of domain 4 (d4) of the cholesterol-dependent cytolysins (cdcs) mediate the binding of the cdc monomers to cholesterol-rich cell membranes. but intermedilysin (ily), from streptococcus intermedius, does not bind to cholesterol-rich membranes unless they contain the human protein cd59. this observation suggested that the d4 loops, which include loops l1-l3 and the undecapeptide, of ily were no longer required for its cell bind ...200717412689
conformational changes that effect oligomerization and initiate pore formation are triggered throughout perfringolysin o upon binding to cholesterol.pore formation by the cholesterol-dependent cytolysins (cdcs) requires the presence of cholesterol in the target membrane. cholesterol was long thought to be the cellular receptor for these toxins, but not all cdcs require cholesterol for binding. intermedilysin, secreted by streptococcus intermedius, only binds to membranes containing the human protein cd59 but forms pores only if the membrane contains sufficient cholesterol. in contrast, perfringolysin o (pfo), secreted by clostridium perfring ...200717553799
