degradation of poly(3-hydroxybutyrate) by poly(3-hydroxybutyrate) depolymerase from alcaligenes faecalis t1.the extracellular poly(3-hydroxybutyrate) depolymerase purified from alcaligenes faecalis t1 has two disulfide bonds, one of which appears to be necessary for the full enzyme activity. this depolymerase hydrolyzed not only hydrophobic poly(3-hydroxybutyrate) but also water-soluble trimer and larger oligomers of d-(-)-3-hydroxybutyrate, regardless of their solubilities in water. kinetic analyses with oligomers of various sizes indicated that the substrate cleaving site of the enzyme consisted of ...19863942778
mechanism of alpha-amino-epsilon-caprolactam racemase reaction.alpha-amino-epsilon-caprolactam racemase catalyzes the exchange of the alpha-hydrogen of the substrate with deuterium during racemization in deuterium oxide. the rate of the hydrogen exchange measured by 1h nmr is lower than that of racemization in deuterium oxide for both the enantiomers. both the enantiomers of alpha-amino-epsilon-caprolactam show an overshoot of the optical rotation during the enzymatic racemization in deuterium oxide (but not in water). this phenomenon may be attributable to ...19863955003

comparison of cross-staining reactions by pseudomonas spp. and fluorescein-labeled polyclonal and monoclonal antibodies directed against legionella pneumophila.commercially prepared polyclonal antisera to legionella pneumophila are known to cross-react with organisms of the genus pseudomonas. to determine whether a commercially available monoclonal antibody reagent specific for l. pneumophila would also cross-react with pseudomonads, a two-laboratory study was undertaken to test both monoclonal and polyclonal reagents against 33 isolates of pseudomonas spp., including 25 pseudomonas aeruginosa, 4 p. putida, 2 p. maltophilia, 1 p. fluorescens, and 1 p. ...19863958153
investigation of cadmium resistance in an alcaligenes sp.the mechanisms of metal resistance of a cadmium-resistant alcaligenes sp. were studied. growth in a defined medium was unaffected by cadmium at concentrations up to 0.1 mm, while at concentrations up to 2.5 mm, growth occurred after an extended lag phase. the increase in length of the lag phase was abolished by repeated subculturing at these higher concentrations. however, subculture in the absence of cadmium reversed the adaptation process. plasmid dna was not detected in adapted cells, suggest ...19863963810
studies on microorganisms secured from different organs of slaughtered sheep with special reference to the microbial load in certain muscles.a total of 312 samples collected from different organs and muscles of dead, disabled, diseased, dying, and normal carcasses of sheep were examined bacteriologically to determine the different species of microorganisms as to their public health significance. in addition, the aerobic plate count (apc) and most probable number (mpn) coliform/g were determined of musculus biceps brachii, musculus quadriceps femoris, and musculus longissimus dorsi. the mean bacterial load was higher in summer than in ...19863964203
properties and electron transfer specificity of copper proteins from the denitrifier "achromobacter cycloclastes".a blue copper protein (mr 12,000) was purified from cells of "achromobacter cycloclastes" grown as a denitrifier. when reduced, the blue copper protein transferred electrons to the copper protein nitrite reductase purified from the same cells, whereas a variety of cytochromes from denitrifiers failed to do so. inclusion of a protease inhibitor, phenylmethylsulfonyl fluoride, in the buffers employed during preparation yielded purified blue copper protein with 18 more amino acid residues and two t ...19863700338
identification of oxidase-positive, glucose-negative, motile species of nonfermentative bacilli.motile and glucose-negative nonfermentative bacilli are difficult to identify by commonly used procedures. we found eight features (reduction of nitrate and nitrite, acidification of fructose, and alkalinization of acetamide, arginine, histidine, saccharate, and urea) that identified 83% of 143 strains. these, along with additional biochemical tests, effected identification of 93% of the strains. hence, only a minority of these strains required examination by microscopy (for type of flagellation ...19863711277

antitumor polysaccharide-induced tumor-regressing factor in the serum of tumor-bearing mice: purification and characterization.marked tumor-regressing activity was induced in the serum of s180 tumor-bearing mice by injection of an antitumor polysaccharide, cm-tak [carboxymethylated beta(1-3)glucan]. maximal activity was induced 7-14 days after the tumor transplantation and 10-12 h after cm-tak treatment. a quantitative assay for the activity was established on the basis of the initial decrease in the number of the tumor cells within 24 h. the factor with tumor-regressing activity was purified 10,000-fold by the series o ...19863723139
[aniline oxidation by microorganisms in chemostat cultivation].under the conditions of chemostat cultivation, a mixed culture of microorganisms oxidized aniline at a gradually increasing concentration (up to 2.5 g/litre) as a sole source of carbon and nitrogen. the specific rate of aniline oxidation was as high as 160 mg per 1 g of dry biomass when aniline concentration in the growth medium was 2.5 g/litre. as aniline concentration in the growth medium was gradually raised, selection of the microorganisms took place and the number of the strains decreased f ...19863724566
the efficacy of a temperature-sensitive mutant vaccine against northwest arkansas isolates of alcaligenes faecalis.the efficacy of a commercially produced temperature-sensitive mutant alcaligenes faecalis vaccine was evaluated in turkeys contact-challenged with one of three strains of a. faecalis. in the vaccinated control group, the vaccine strain of a. faecalis colonized the nasal turbinates but not the trachea, caused no clinical signs of turkey coryza, and induced humoral antibodies. in the vaccinated challenged groups, the vaccine reduced both the severity of lesions and the number of birds exhibiting l ...19863729855
tracheal mucus transport rate in normal turkeys and in turkeys infected with bordetella avium (alcaligenes faecalis).using the radiopharmaceutical 99mtechnetium-sulfur colloid, the tracheal mucus transport rate (tmtr) was measured in healthy unanesthetized turkeys and in turkeys infected with bordetella avium. the tmtr of uninfected turkeys was 35.6 +/- 14.4 cm/min. the tmtr of b. avium-infected turkeys was normal on days 0 through 14 postexposure (pe), despite heavy bacterial colonization of the tracheal epithelium. on day 21 pe, the tmtr of b. avium-infected turkeys was significantly depressed (p less than o ...19863729858
contribution to the taxonomy of the turkey coryza agent: cellular fatty acid analysis of the bacterium.gas-liquid chromatography was used to analyze bacterial cellular fatty acids to elucidate the relatedness of the turkey coryza (tc) bacterium to alcaligenes spp., bordetella spp., and other gram-negative bacteria. the results indicated that the tc bacterium is not closely related to alcaligenes faecalis or any of the reference strains of alcaligenes and bordetella studied. most urease-positive bacterial isolates obtained from the upper respiratory tract of turkeys were identified as bordetella b ...19863729861
transient kinetics of reduction of blue copper proteins by free flavin and flavodoxin semiquinones.rate constants have been determined for the electron-transfer reactions between reduced free flavins and flavodoxin semiquinone and several blue copper proteins. correlations between these values and redox potentials demonstrate that spinach plastocyanin, pseudomonas aeruginosa azurin, alcaligenes sp. azurin, and alcaligenes sp. nitrite reductase have the same intrinsic reactivities toward free flavins, whereas stellacyanin is more reactive (3.3 times) and laccase considerably less reactive (app ...19863730365
quantitative and qualitative studies of gut flora in striped bass from estuarine and coastal marine environments.examination of the intestinal contents of 130 striped bass (morone saxatilis) collected from the hudson river and long island sound during may to october 1981 showed that opportunistic fish pathogens--especially aeromonas hydrophila--predominated in samples from both locations. other isolates from both groups of striped bass included vibrio, pseudomonads, flavo-bacteria, alcaligenes, and enterics. small numbers of micrococcus, bacillus, corynebacterium, and acinetobacter were also isolated. tota ...19863735580

[amino acid metabolism by thin layer chromatography for the differentiation of bordetella and alcaligenes species].the amino acid metabolism of bordetella and alcaligenes strains was investigated for differentiation purposes. all species investigated reacted differently with a number of amino acids. the genus bordetella could be differentiated from that of alcaligenes by its inability to metabolize l-lysine, l-arginine and d-histidine. the recently described species bordetella avium, as well as achromobacter xylosoxidans (yabuuchi), considered as alcaligenes denitrificans subsp. xylosoxidans, presented thems ...19863788350
degradation of 1,4-dichlorobenzene by alcaligenes sp. strain a175.an organism, identified as an alcaligenes sp., was isolated from an enrichment culture in which 1,4-dichlorobenzene served as the sole carbon and energy source. during growth with 1,4-dichlorobenzene in pure culture, stoichiometric amounts of chloride were released. growth experiments and oxygen uptake rates with other chlorinated aromatic compounds revealed a high degree of specificity of the initial dioxygenase. cis-1,2-dihydroxycyclohexa-3,5-diene oxidoreductase and 1,2-pyrocatechase, but not ...19863789724
oxygenase properties of the (4-hydroxybenzoyl)methanol-cleavage enzyme from an alcaligenes sp.studies using h2(18)o and 18o2 demonstrated that, in the cleavage of (4-hydroxybenzoyl)methanol to 4-hydroxybenzoic and formic acids by an enzyme from an alicaligenes sp., oxygen is incorporated into both products from dioxygen and not from water. the enzyme is, therefore, an oxygenase.19863814084

plasmid dna of virulent alcaligenes faecalis.alcaligenes faecalis strains originating from chickens and from epizootics of coryza in turkeys were screened for antibiotic susceptibility and for the presence of plasmid dna. seven of 35 strains contained plasmid dna ranging in size from 10.5 to approximately 32 megadaltons. all of the strains isolated from turkeys were virulent in turkey poults, but only the plasmid-containing strains were resistant to sulfonamides and streptomycin. four of the plasmid-containing strains were also resistant t ...19863008603
activation and active sites of nickel-containing hydrogenases.hydrogenases that contain nickel and iron-sulphur clusters also have a regulatory mechanism, by which exposure to oxidants such as oxygen prevents their reaction with hydrogen. treatment with reducing agents then causes reactivation. in some hydrogenases from desulfovibrio species, there is evidence that there are at least two different deactivated states, which differ in their rates of reductive reactivation. the membrane-bound hydrogenase of d. desulfuricans, norway strain, the periplasmic hyd ...19863015251
purification and some properties of cytochrome c' from a strain of achromobacter xylosoxidans.cytochrome c' was crystallized from achromobacter xylosoxidans gifu 543. the cytochrome was a basic protein and its molecular weight was 28,000. the pyridine ferrohemochrome showed absorption peaks at 415, 521, and 551 nm. the absorption spectra of the oxidized and reduced forms at neutral ph were almost the same as those of other cytochromes c' reported already. the reduced cytochrome c' reacted with co and no, and the no complex showed a characteristic absorption spectrum. the midpoint redox p ...19863017925
[peptide inhibitors of a bacterial collagenase].the properties of the cleavage site of collagen by collagenases remain discussed. the authors report their studies on the active site of the collagenase of a bacteria, achromobacter, based on two types of methods. the first type uses synthetic substrates, the second one enzymatic inhibitors. the results of both methods are reviewed. the inhibitors seem more sensitive than substrates to study the relation between structure and activity of the enzyme.19863019185
