predicted structure and possible ionmotive mechanism of the sodium-linked nadh-ubiquinone oxidoreductase of vibrio alginolyticus.two groups have now published sequences of the six genes contained in the operon coding for the sodium-linked nadh-ubiquinone oxidoreductase of vibrio alginolyticus. sequence analyses indicate that this enzyme is unrelated to other known respiratory nadh dehydrogenases. a search for cofactor motifs suggests that the enzyme contains only one fad, a ferredoxin-type iron sulphur centre, and the nadh-binding site. these are all located on nqrf, a subunit that can be recognized as a new member of a l ...19957498478
a gene encoding a thermophilic alkaline serine proteinase from thermus sp. strain rt41a and its expression in escherichia coli.the extreme thermophile thermus sp. strain rt41a produces an extracellular alkaline serine proteinase during growth. this enzyme is stable for more than 24 h at 70 degrees c and has a ph optimum of 8.0. the proteinase gene was identified using primers designed to amplify a region between two highly conserved amino acid motifs in subtilisin-like proteinases and the pcr product was used to identify a genomic fragment containing the gene. the amino acid sequence deduced from the rt41a gene containe ...19957551039
reexamination of tetrodotoxin production by bacteria.vibrio alginolyticus has been reported as a good producer of tetrodotoxin (ttx), but the toxin extracted from this bacterium did not react to the monoclonal antibody against ttx. surprisingly, chromatographic analyses detected high ttx peaks for polypeptone and yeast extracts used as medium materials, which were, as expected, all negative by the mouse bioassay. these results may require us to revise the bacterial production of ttx.19957574655

chemotactic responses to an attractant and a repellent by the polar and lateral flagellar systems of vibrio alginolyticus.chemotactic responses in vibrio alginolyticus, which has lateral and polar flagellar systems in one cell, were investigated. a lateral-flagella-defective (pof+ laf-) mutant, which has only a polar flagellum, usually swam forward by the pushing action of its flagellum and occasionally changed direction by backward swimming. when the repellent phenol was added, pof+ laf- cells moved frequently forward and backward (tumbling state). the tumbling was derived from the frequent changing between counte ...19968885393
isolation and characterization of vibrio alginolyticus isolated from diseased kuruma prawn, penaeus japonicus.outbreaks of serious mortality among cultured kuruma prawns (penaeus japonicus) with white spotted syndrome in the carapace occurred in the summer of 1993 in i-lan, taiwan. a swarming bacterium, strain swy, was isolated from the hepatopancreas of the moribund prawns using tryptic soy agar supplemented with 1% nacl and/or thiosulphate citrate bile salt sucrose agar. this strain was characterized and identified as vibrio alginolyticus on the basis of a number of biochemical tests. the swy strain w ...19968936369
cloning and sequencing of novel genes from vibrio alginolyticus that support the growth of k+ uptake-deficient mutant of escherichia coli.novel genes that functionally complement the growth of k+ uptake-deficient mutant strain of escherichia coli tk420 have been cloned from the marine bacterium vibrio alginolyticus. the nucleotide sequence revealed three open reading frames. the second gene was homologous to proc gene and allowed the growth of proc-defective mutant strain of e. coli chi 342 in the absence of proline. the first and third genes, but not proc, were required for the growth of tk420 in a synthetic medium containing 10 ...19968982386
alkaline serine protease is an exotoxin of vibrio alginolyticus in kuruma prawn, penaeus extracellular lethal toxin produced by vibrio alginolyticus strain swy originally isolated from diseased kuruma prawn(penaeus japonicus) was partially purified by fast protein liquid chromatography with hydrophobic interaction (phenyl sepharose hig hperformance) chromatography and gel filtration columns. the toxin is an alkaline serine protease, inhibited by phenyl-methylsulfonyl fluoride (pmsf),and showed maximal activity at ph 10, having a molecular weight of about 33kda estimated by sds-pa ...19979003588
vibrio alginolyticus mutants resistant to phenamil, a specific inhibitor of the sodium-driven flagellar motor.the polar flagella of vibrio alginolyticus are driven by sodium motive force and those motors are specifically and strongly inhibited by phenamil, an amiloride analog that is thought to interact with a sodium channel of the flagellar motor. to study the sodium ion coupling site, we isolated motility mutants resistant to phenamil and named the phenotype mpa(r) for motility resistant to phenamil. the motility of the wild-type (mpa(s)) was inhibited by 50 microm phenamil, whereas mpa(r) strains wer ...19979018045
genetic defect of the sodium pump-defective mutant nap-1 from the marine vibrio alginolyticus.the marine bacterium, vibrio alginolyticus, has a respiratory chain-linked na(+)-translocating nadh-quinone reductase (nqr). among several mutant cells defective in na+ pump activity, nap1 was a very stable mutant and a spontaneous revertant could not be isolated from nap1. using genetic information from the recently sequenced nqr operon, the genetic defects in nap1 were examined, and the sodium pump-defective mutant nap1 was found to be caused by the insertion of a 1.2 kbp dna fragment into the ...19979043633
membrane localization, topology, and mutual stabilization of the rnfabc gene products in rhodobacter capsulatus and implications for a new family of energy-coupling nadh oxidoreductases.the rnf genes in rhodobacter capsulatus are unique nitrogen fixation genes that encode potential membrane proteins (rnfa, rnfd, and rnfe) and potential iron-sulfur proteins (rnfb and rnfc). in this study, we first analyzed the localization and topology of the rnfa, rnfb, and rnfc proteins. by activity and immunoblot analysis of expression of translational fusions to escherichia coli alkaline phosphatase, rnfa protein was shown to span the chromatophore membrane with its odd-numbered hydrophilic ...19979154934
the na+/e- stoichiometry of the na+-motive nadh:quinone oxidoreductase in vibrio alginolyticus.a method is proposed to estimate the stoichiometries of primary na+-pumps in intact bacterial cells. it is based on technique when the h+/e- stoichiometry is measured in the presence of protophorous uncoupler and in the absence of penetrating ions other than h+. under these conditions, the h+ influx discharges membrane potential generated by the na+ pump so the na+/e- and h+/e- ratios become equal. using this approach it is shown that the na+/e- ratio for the na+-motive nadh:quinone oxidoreducta ...19979224712
putative channel components for the fast-rotating sodium-driven flagellar motor of a marine bacterium.the polar flagellum of vibrio alginolyticus rotates remarkably fast (up to 1,700 revolutions per second) by using a motor driven by sodium ions. two genes, motx and moty, for the sodium-driven flagellar motor have been identified in marine bacteria, vibrio parahaemolyticus and v. alginolyticus. they have no similarity to the genes for proton-driven motors, mota and motb, whose products constitute a proton channel. motx was proposed to be a component of a sodium channel. here we identified additi ...19979260952
effects of extracellular products of vibrio alginolyticus on penaeid prawn plasma components.the effects of both crude extracellular products (ecp) and a partially purified protease of vibrio alginolyticus on the plasma components of kuruma prawn (penaeus japonicus) and tiger prawn (p. monodon) were studied using crossed immunoelectrophoresis (cie). a component of the plasma, tentatively identified as coagulogen, apparently disappeared after incubation with the ecp, while the amount of a component tentatively identified as haemocyanin decreased. the coagulogen and an unknown component ( ...19979281857
h+-pumping atpase has little stimulatory effect on in vitro translocation of a model protein into vibrio alginolyticus inside-out membrane vesicles.we studied the effect of a h+ electrochemical potential generated by f1f0 atpase on in vitro translocation of a model protein into vibrio alginolyticus inside-out membrane vesicles. the f1f0 atpase of v. alginolyticus catalyzed the pumping of h+ coupled to atp hydrolysis as measured in fluorescence quenching experiments. consequently, this enzyme leads to the generation of a h+ electrochemical potential. the h+ electrochemical potential generated by f1f0 atpase was completely abolished by 30 mic ...19979339889
cloning of a vibrio alginolyticus rpon gene that is required for polar flagellar formation.a fragment of dna was cloned which complemented a polar flagellum-defective (pof) mutation of vibrio alginolyticus. the fragment contained two complete and two partial open reading frames (orfs) (orf2 and -3 and orf1 and -4, respectively). the presumed product of orf2 has an amino acid sequence with a high degree of similarity to that of rpon, which is an alternative sigma factor (sigma54) for other microorganisms. the other orfs are also homologous to the genes adjacent to other rpon genes. del ...19979352939
generation of reactive oxygen species by raphidophycean phytoplankton.chattonella marina, a raphidophycean flagellate, is one of the most toxic red tide phytoplankton and causes severe damage to fish farming. recent studies demonstrated that chattonella sp. generates superoxide (o2-), hydrogen peroxide (h2o2), and hydroxyl radicals (.oh), which may be responsible for the toxicity of c. marina. in this study, we found the other raphidophycean flagellates such as heterosigma akashiwo, olisthodiscus luteus, and fibrocapsa japonica also produce o2- and h2o2 under norm ...19979362113
the na+-translocating nadh:ubiquinone oxidoreductase from vibrio alginolyticus--redox states of the fad prosthetic group and mechanism of ag+ inhibition.the fad prosthetic group of the na+-motive nadh:ubiquinone oxidoreductase (na+-nqr) from vibrio alginolyticus was investigated by ultraviolet-visible and fluorescence spectroscopy. the reduction of na+-nqr by excess nadh in the presence of 6-13 microm o2 resulted in the formation of the blue flavosemiquinone radical. if the concentration of dioxygen was further reduced to 0.1 microm o2, neither the reduction of na+-nqr by nadh nor its reoxidation with ubiquinone-1 (q-1) yielded a stable flavosem ...19979395325
evaluation of vitek gni+ and becton dickinson microbiology systems crystal e/nf identification systems for identification of members of the family enterobacteriaceae and other gram-negative, glucose-fermenting and non-glucose-fermenting bacilli.we evaluated the vitek gni+ and becton dickinson crystal e/nf identification systems for their ability to accurately identify 619 and 626 strains, respectively, of members of the family enterobacteriaceae and other glucose-fermenting and non-glucose-fermenting gram-negative rods. all strains tested were taken from a stock collection and passed three times on 5% sheep blood agar prior to testing. these strains represented a more rigorous challenge to both systems than one resulting from the testi ...19979399532

cloning and nucleotide sequence of the gyrb gene of vibrio parahaemolyticus and its application in detection of this pathogen in shrimp.because biochemical testing and 16s rrna sequence analysis have proven inadequate for the differentiation of vibrio parahaemolyticus from closely related species, we employed the gyrase b gene (gyrb) as a molecular diagnostic probe. the gyrb genes of v. parahaemolyticus and closely related vibrio alginolyticus were cloned and sequenced. oligonucleotide pcr primers were designed for the amplification of a 285-bp fragment from within gyrb specific for v. parahaemolyticus. these primers recognized ...19989464408
attachment of vibrio alginolyticus to glass surfaces is dependent on swimming speed.the attachment of vibrio alginolyticus to glass surfaces was investigated with special reference to the swimming speed due to the polar flagellum. this bacterium has two types of flagella, i.e., one polar flagellum and numerous lateral flagella. the mutant ym4, which possesses only the polar flagellum, showed much faster attachment than the mutant ym18, which does not possess flagella, indicating that the polar flagellum plays an important role. the attachment of ym4 was dependent on na+ concent ...19989473049
identification of six subunits constituting na+-translocating nadh-quinone reductase from the marine vibrio alginolyticus.we previously reported that the purified na+-translocating nadh-quinone reductase (nqr) from the marine vibrio alginolyticus is composed of three major subunits, alpha, beta and gamma. nqr operon was sequenced and was found to be composed of 6 structural genes. among these genes, nqr1, nqr3 and nqr6 were identified to code for alpha-, gamma- and beta-subunits, respectively. the protein products from nqr2, nqr4 and nqr5, however, were not reported. the sequence data predicted that these three pro ...19989490015
overexpression in escherichia coli of the rnf genes from rhodobacter capsulatus--characterization of two membrane-bound iron-sulfur proteins.the rnf genes of rhodobacter capsulatus, essential for nitrogen fixation, are thought to encode a system for electron transport to nitrogenase. in the present study, we have attempted to overexpress the rnf genes in escherichia coli to investigate the molecular properties of the corresponding proteins. corrections were made to the published dna sequence of the rnf operon, resulting in the identification of two genes, rnfg and rnfh. the rnfabcdgeh operon thus comprises seven genes and shows simil ...19989492268
polarizing response of human polymorphonuclear cells to vibrio cholerae.leucocytes have the capacity to respond to chemotactic factors by becoming morphologically and functionally polarized and this method has been found to be suitable for measurement of chemotaxis. this work evaluates the effect of whole vibrio cholerae biotype el tor polarization of human polymorphonuclear (pmn) cells in comparison with strains of v. cholerae nag, vibrio alginolyticus and salmonella typhi. v. cholerae o1 induced, at different cell/bacteria ratios, a significant increase in the per ...19979503599
topological study of vibrio alginolyticus nhab na+/h+ antiporter using gene fusions in escherichia coli cells.nhab, an na+/h+ antiporter, of vibrio alginolyticus is a 528-amino-acid protein. hydropathy profile-based computer analysis predicted that the nhab might contain up to 13 membrane-spanning domains. to examine this hypothesis, we applied the phoa fusion method to the cloned nhab gene. eighteen plasmid-borne nhab-phoa fusion genes were constructed in escherichia coli cells and the alkaline phosphatase activity and expression level of the fusion proteins analyzed. these results and the results obta ...19989518558

chemotaxis of pathogenic vibrio strains towards mucus surfaces of gilt-head sea bream (sparus aurata l.).vibrio anguillarum and vibrio alginolyticus exhibited significant adhesion to and chemotactic abilities towards mucus collected from the skin, gills, and intestine of gilt-head sea bream. quadratic polynomial models for chemotaxis designed to estimate the influence of temperature demonstrated a differential bacterial chemotaxis depending of the source of the mucus, with the chemotaxis towards intestinal mucus being the least influenced.19989575135
flagellin-containing membrane vesicles excreted from vibrio alginolyticus mutants lacking a polar-flagellar filament.polar flagellum-defective mutants (pof- laf-) have been isolated from a lateral flagella-defective mutant (pof+ laf-). among these pof- laf- mutants, polar-filamentless mutants, which have the hook structure but not the filament, were identified by electron microscopy. their hooks were covered with a sheath structure which is contiguous to the outer membrane. the filament proteins, flagellins, were shed into the culture medium of these mutants. these flagellins could be sedimented by high-speed ...19989604007
ktrab, a new type of bacterial k(+)-uptake system from vibrio alginolyticus.vibrio alginolyticus contained two adjacent genes, ktra and ktrb, which encode a new type of bacterial k(+)-uptake system. ktra and ktrb are peripheral and integral membrane proteins, respectively. six of the nine sequenced bacterial genomes contain homologs to both ktra and ktrb, suggesting that ktrab is widespread.19989642210
cloning of the trkah gene cluster and characterization of the trk k(+)-uptake system of vibrio alginolyticus.k(+)-uptake genes of vibrio alginolyticus were identified by cloning chromosomal dna fragments of this organism into plasmids, followed by electroporation and selection for growth at low k+ concentrations of cells of an escherichia coli strain defective in k+ uptake. a 4.1 kb dna fragment contained a cluster of three orfs on the same dna strand: the previously identified trka gene, a gene similar to e. coli trkh (v. alginolyticus trkh) and a new gene, orf1, whose function is not clear. products ...19989720051
cloning and expression of the seca gene of a marine bacterium, vibrio alginolyticus, and analysis of its function in escherichia coli.we report the cloning, sequencing and functional characterization of the seca gene of a marine bacterium, vibrio alginolyticus, which has been suggested to utilize atp and the sodium motive force for protein translocation. oligodeoxynucleotides corresponding to highly conserved regions of escherichia coli seca located in the high affinity atp binding site were utilized as pcr primers to clone the seca gene of v. alginolyticus. it was shown to encode a 103.3-kda protein. the deduced amino acid se ...19989729436
necrotizing fasciitis due to vibrio alginolyticus following an injury inflicted by a stingray.we describe the case of a patient with necrotizing fasciitis due to vibrio alginolyticus in a patient with cirrhosis following an injury inflicted by a stingray. the patient was successfully treated with aggressive surgical debridement and a combination of ciprofloxacin and amoxicillin-clavulanate. cases of invasive v. alginolyticus reported in the literature were reviewed.19989730311
investigation of the 1994-5 ukrainian vibrio cholerae epidemic using molecular methods.thirty-seven vibrio cholerae and four non-cholera vibrio isolates from ukraine, including strains from the epidemic of 1994-5, were analysed by molecular methods. results from pfge and ribotyping indicated that all ukrainian toxigenic v. cholerae were closely related to each other and to an isolate from a patient from pakistan. a non-toxigenic river water strain obtained during the height of the epidemic was more distantly related to these v. cholerae strains, while the vibrio parahaemolyticus i ...19989747752
molecular cloning and functional characterization of sece of a marine bacterium, vibrio alginolyticus.sece is an essential membrane component of the escherichia coli protein translocation machinery, which utilizes atp and the proton motive force as energy sources. the sece homologue of marine vibrio alginolyticus, in which protein translocation requires atp and the sodium motive force, was cloned, sequenced, and characterized. unlike most sece homologues found in various organisms, sece of v. alginolyticus (va-sece) possesses three transmembrane sequences like e. coli sece (ec-sece) and compleme ...19989791006
pathogenicity of vibrio alginolyticus for cultured gilt-head sea bream (sparus aurata l.).the in vivo and in vitro pathogenic activities of whole cells and extracellular products of vibrio alginolyticus for cultured gilt-head sea bream were evaluated. the 50% lethal doses ranged from 5.4 x 10(4) to 1.0 x 10(6) cfu/g of body weight. the strains examined had the ability to adhere to skin, gill, and intestinal mucus of sea bream and to cultured cells of a chinook salmon embryo cell line. in addition, the in vitro ability of v. alginolyticus to adhere to mucus and skin cells of sea bream ...19989797276

neurotoxic effect on two fish species and a pc12 cell line of the supernate of vibrio alginolyticus and vibrio anguillarum.the biological effects of supernates obtained from different strains of vibrio alginolyticus and vibrio anguillarum isolated from diseased fish have been studied by inoculation on two fish species, eel and rainbow trout, and two fish cell lines. these supernates possess neuroexcitatory properties, and so, when they are injected into both fish species, they trigger convulsions, wriggling, contortive swimming and respiratory arrest coupled with increased respiratory reflex. furthermore, after the ...19989810622
mechanosensitive channel functions to alleviate the cell lysis of marine bacterium, vibrio alginolyticus, by osmotic downshock.the mechanosensitive channel with large conductance of escherichia coli is the first to be cloned among stretch-activated channels. although its activity was characterized by a patch clamp method, a physiological role of the channel has not been proved. the marine bacterium, vibrio alginolyticus, is sensitive to osmotic stress and cell lysis occurs under osmotic downshock. we introduced an mscl gene into vibrio alginolyticus, and the mechanosensitive channel with large conductance functions was ...199910050752
the polar flagellar motor of vibrio cholerae is driven by an na+ motive force.vibrio cholerae is a highly motile bacterium which possesses a single polar flagellum as a locomotion organelle. motility is thought to be an important factor for the virulence of v. cholerae. the genome sequencing project of this organism is in progress, and the genes that are highly homologous to the essential genes of the na+-driven polar flagellar motor of vibrio alginolyticus were found in the genome database of v. cholerae. the energy source of its flagellar motor was investigated. we exam ...199910074090
identification of vibrio parahaemolyticus strains at the species level by pcr targeted to the toxr gene.the dna colony hybridization test with the polynucleotide probe for vibrio parahaemolyticus toxr gene was performed. all 373 strains of v. parahaemolyticus gave positive results, and the strains belonging to four other vibrio species including vibrio alginolyticus gave weakly positive results, suggesting that toxr sequence variation may reflect the phylogenetic relationships of vibrio species. we then established a toxr-targeted pcr protocol for the specific detection of v. parahaemolyticus.199910074546
